Analysis of heat generation during the production of drug nanosuspensions in a wet stirred media mill

Gulenay Guner et al.

Summary: This study investigated the impact of stirrer speed, bead loading, and bead size on the temperature evolution during the milling process. The results showed that stirrer speed had the most significant impact on temperature increase, followed by bead loading and bead size. The study also established a power number correlation to calculate power and determine the heat generation rate.

Mechanistic Modeling of Wet Stirred Media Milling for Production of Drug Nanosuspensions

E. Bilgili et al.

Summary: Drug nanocrystals are widely used for enhancing the bioavailability of poorly water-soluble drugs in the pharmaceutical industry, with wet stirred media milling being the main process. Despite numerous review papers in the field, modeling of WSMM has not been fully covered, and more mechanistic approaches could provide a fundamental understanding for pharmaceutical engineers. Advancements in microhydrodynamic modeling were highlighted, with potential research directions including more accurate simulations and model couplings.
