Dysfunction of the glutamatergic photoreceptor synapse in the P301S mouse model of tauopathy

Summary: Tauopathies, including Alzheimer's disease, lead to loss of retinal ganglion cells and the deposition of amyloid and phosphorylated tau protein. In the murine P301S tauopathy model, visual impairments were observed at 6 months, indicating the functional impact of these histopathological alterations. Delayed visual signals to the cortex and retina were observed at 6 and 9 months, respectively. Surprisingly, the retinal output signal showed opposite delays at the light onset and offset, suggesting a dysfunction of the cone photoreceptor synapse. This discovery highlights the potential value of the retina in the diagnostic assessment and evaluation of therapies for Alzheimer's disease and other tauopathies.
