Emerging role of ferroptosis in breast cancer: New dawn for overcoming tumor progression

Summary: Breast cancer is a significant threat to women's health, and ferroptosis may offer a new direction for breast cancer treatment. However, the complete appearance of ferroptosis in breast cancer and its potential combination with traditional anti-breast cancer drugs remain uncertain. This review explores the roles of various factors associated with ferroptosis in breast cancer, provides evidence of ferroptosis induction by traditional drugs and elimination of breast cancer cells by ferroptosis inducers, and discusses the prospects and potential obstacles of using ferroptosis inducers in breast cancer therapy.

Cancer stem cell regulated phenotypic plasticity protects metastasized cancer cells from ferroptosis

Summary: The study reveals the role of breast cancer stem cell (BCSC) secretome in regulating the stem cell population, tumor-initiating capacity, and metastasis. DKK1 is identified as a key factor secreted by BCSCs that promotes differentiation and metastatic outgrowth. DKK1 increases the expression of SLC7A11 to protect metastasizing cancer cells from ferroptosis. Combination therapy with a ferroptosis inducer and a DKK1 inhibitor shows synergistic effects in reducing metastasis.

Summary: Cuproptosis is a newly recognized regulated cell death mechanism that shows promise in cancer treatment. A novel copper/iron hybrid hollow metal organic framework (HaMOF) is developed for synergistic anticancer therapy by amplifying oxidative stress and disrupting copper/iron metabolism. The bimetallic hollow nanoplatform induces potent ferroptosis and cuproptosis, and efficiently suppresses tumor growth.
