Triterpenes from Pholiota populnea as Cytotoxic Agents and Chemosensitizers to Overcome Multidrug Resistance of Cancer Cells

Morteza Yazdani et al.

Summary: A detailed mycochemical analysis of Pholiota populnea led to the isolation of four new lanostane diesters and one acyclic triterpene. These compounds showed cytotoxic activity against colon adenocarcinoma and non-tumoral cell lines, with ergosterol showing substantial cytotoxic activity against all cell lines. The study also revealed that some of the isolated compounds have the capacity to inhibit the efflux-pump overexpressed in resistant cancer cells, and certain combinations of compounds showed strong synergistic effects with doxorubicin. These findings highlight the potential of P. populnea as a source of new triterpenes with chemosensitizing activity on cancer cells.

Triterpenoids with modified A-ring as modulators of P-gp-dependent drug-resistance in cancer cells

Ekaterina Yu Rybalkina et al.

Summary: The semi-synthetic A-cycle modified triterpenic derivatives showed cytotoxicity against tumor cells and were similar to classical inhibitors in their action mechanism. Combining the modified triterpenic derivatives with doxorubicin improved the cytotoxic effect of the therapeutic drug against resistant tumor cells.

Exploration of Benzenesulfonamide-Bearing Imidazole Derivatives Activity in Triple-Negative Breast Cancer and Melanoma 2D and 3D Cell Cultures

Benas Balandis et al.

Summary: A series of new imidazole derivatives incorporating a benzenesulfonamide moiety were synthesized and evaluated for cytotoxicity against human triple-negative breast cancer and malignant melanoma cells. Compounds containing 4-chloro and 3,4-dichlorosubstituents in the benzene ring and 2-ethylthio and 3-ethyl groups in the imidazole ring were determined as the most active. The most cytotoxic compound showed EC50 values of 27.8 & PLUSMN; 2.8 μM against IGR39 cells and 20.5 & PLUSMN; 3.6 μM against MDA-MB-231 cells.

Dietary Polyphenols: Promising Adjuvants for Colorectal Cancer Therapies

Laura Bracci et al.

Summary: Colorectal cancer is a major global cause of death, and the current medical treatments are not sufficient to control metastatic disease. Combination therapy involving cytotoxic treatments and non-toxic natural compounds is seen as a promising more effective and safer alternative. Polyphenols found in fruit and vegetables have shown potential for enhancing colorectal cancer therapy, but further clinical studies are needed to definitively prove their value.

Betulinic Acid-Doxorubicin-Drug Combination Induced Apoptotic Death via ROS Stimulation in a Relapsed AML MOLM-13 Cell Model

Milan Vu et al.

Summary: The study revealed a synergistic growth inhibitory effect of the combination of betulinic acid and doxorubicin on MOLM-13 cells in AML. The combination treatment caused more cells to reside in the late apoptotic stage through elevated ROS generation, potentially disrupting mitochondrial activity in MOLM-13 cell death.
