AlphaFold2: A Role for Disordered Protein/Region Prediction?

Carter J. Wilson et al.

Summary: The development of AlphaFold2 has brought a paradigm shift in structural biology and has shown excellent performance in predicting disordered regions. The accuracy of the constructed disorder predictor from AlphaFold2 structures varies, with the naive assumption of assigning residues to ordered or disordered based on helices, strands, and H-bond stabilized turns leading to a significant overestimation of disorder. The predicted local distance difference test (pLDDT) provides a reliable measure of residue-wise disorder and shows an interesting relationship with secondary structure.

Summary: Components of the extracellular matrix (ECM) are found to be associated with complement activation and inflammation, while the proteins FHR1 and FHR5 are implicated in renal and eye diseases. It has been discovered that FHR1 and FHR5 can bind to specific ECM components, affecting the activity of complement inhibitor FH and promoting complement activation.

Single-sequence protein structure prediction using a language model and deep learning

Ratul Chowdhury et al.

Summary: This article reports the development of an end-to-end differentiable recurrent geometric network (RGN) called RGN2 for predicting protein structure using the protein sequence. Compared to AlphaFold2 and RoseTTAFold, RGN2 performs better on orphan proteins and designed protein classes, while achieving a computational speedup of up to 10^6-fold. These findings demonstrate the practical and theoretical advantages of protein language models over multiple sequence alignments (MSAs) in protein structure prediction.

Complement inhibition at the level of C3 or C5: mechanistic reasons for ongoing terminal pathway activity

Marco Mannes et al.

Summary: Use of C5 inhibitor eculizumab to block the terminal complement pathway has revolutionized clinical management of complement-mediated diseases. New findings show that C3/C5 convertases function differently than previously thought, with C3 inhibition not necessarily leading to C5 inhibition. Surface-deposited C4b alone can recruit and activate C5, and only C5 bound on C3b/C4b-decorated surfaces requires surface fixation for activation.

Molecular bases for the association of FHR-1 with atypical hemolytic uremic syndrome and other diseases

Hector Martin Merinero et al.

Summary: Factor H (FH)-related protein 1 (FHR-1) plays a significant role in diseases like atypical hemolytic uremic syndrome (aHUS) by competing with FH in binding to surface-bound C3b to promote complement activation. Abnormally elevated FHR-1/FH activity ratios perpetuate pathological complement dysregulation at complement-activating surfaces.

Deregulation of Factor H by Factor H-Related Protein 1 Depends on Sialylation of Host Surfaces

Summary: The SV-to-LA substitution in the C-terminal regions of FH and FHR-1 diminishes their sialic acid-binding ability, resulting in mild FH deregulation by FHR-1 only on host or host-like surfaces. Conversion of FHR-1 into a sialic acid binder enhances its deregulatory capacity, explaining the pathophysiology of the aHUS-associated FHR-1 SV variant.
