Summary: Autophagy is a homeostatic process that regulates the development of pancreatic ductal adenocarcinoma (PDAC). In a genetically engineered mouse model, we found that blocking autophagy reduced the incidence of PDAC, but did not affect the survival time of animals with tumors. Additionally, the absence of autophagy led to changes in the structure of the pancreas and a decrease in the number of insulin-expressing cells.

Autophagy: A Key Player in Pancreatic Cancer Progression and a Potential Drug Target

Therefore, investigating therapeutic strategies targeting the autophagy pathway is crucial for overcoming chemo-resistance in pancreatic cancer.

These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Cancer cells escape autophagy inhibition via NRF2-induced macropinocytosis

Summary: Although many cancers rely on autophagy for nutrient acquisition, blocking autophagy alone does not effectively inhibit tumor growth. Research has shown that blocking autophagy can lead to pancreatic ductal adenocarcinoma (PDAC) upregulating an alternative nutrient pathway, macropinocytosis (MP), allowing tumor cells to extract nutrients from outside sources. This switch from autophagy to MP, driven by the activation of transcription factor NRF2, presents a potential therapeutic strategy for inhibiting tumor growth and regression.
