Upregulation of exosomal circPLK1 promotes the development of non-small cell lung cancer through the miR-1294/ high mobility group protein A1 axis

Summary: This study found that circPLK1 expression was upregulated in NSCLC patients and its high expression was associated with a poor prognosis. Exosomes derived from NSCLC serum could enhance the migration, invasion, and replication of NSCLC cells while inhibiting apoptosis. The overexpression of circPLK1 also promoted the malignant phenotype of NSCLC cells. Molecular analyses revealed that circPLK1 directly targeted miR-1294 and negatively regulated its activity, resulting in the progression of NSCLC. The findings suggest a potential therapeutic strategy targeting exosomal circPLK1 in NSCLC treatment.

Serum-derived extracellular vesicles facilitate temozolomide resistance in glioblastoma through a HOTAIR-dependent mechanism

Summary: In this study, the researchers found that extracellular vesicles (EVs) transfer a long non-coding RNA (lncRNA) called HOTAIR, which regulates resistance to the chemotherapy drug temozolomide (TMZ) in glioblastoma (GBM). Through in vitro and in vivo experiments, it was demonstrated that HOTAIR promotes tumor growth, increases invasiveness, and enhances resistance to TMZ. In GBM cells, HOTAIR competitively binds to miR-526b-3p, weakening its ability to bind to EVA1 and consequently increasing EVA1 expression. Furthermore, HOTAIR carried by serum-EVs suppresses miR-526b-3p-mediated inhibition of EVA1, promoting tumor growth and TMZ resistance in vivo.

The underlying mechanism involved miR-769-5p targeting CASP9 and promoting the degradation of p53 through the ubiquitin-proteasome pathway.
