PROTAC targeted protein degraders: the past is prologue

Miklos Bekes et al.

Summary: Targeted protein degradation (TPD) is a new therapeutic modality that can tackle disease-causing proteins which are difficult to target with conventional small molecules. PROTAC molecules, utilizing the ubiquitin-proteasome system to degrade target proteins, has achieved clinical proof-of-concept and attracted significant industry activity. Future directions include identifying target classes suitable for TPD, expanding the use of ubiquitin ligases for precision medicine, and extending the modality beyond oncology.

Deubiquitinase-targeting chimeras for targeted protein stabilization

Nathaniel J. Henning et al.

Summary: Many diseases caused by aberrant protein ubiquitination and degradation could benefit from targeted protein stabilization (TPS). In this study, deubiquitinase-targeting chimeras (DUBTACs) were used to stabilize specific proteins degraded in a ubiquitin-dependent manner, leading to therapeutic benefits. Covalent chemoproteomic approaches were used to identify a ligand, EN523, that targeted a specific deubiquitinase. DUBTACs consisting of EN523 and a protein-targeting ligand were shown to stabilize the levels of disease-related proteins, showing promise for TPS.

Cullin-RING Ubiquitin Ligase Regulatory Circuits: A Quarter Century Beyond the F-Box Hypothesis

J. Wade Harper et al.

Summary: Cullin-RING ubiquitin ligases (CRLs) are modular platforms that regulate biological processes by ubiquitylating specific targets. Regulation is achieved through dynamic control of activation and substrate ubiquitylation, and CRLs provide a vast landscape for developing small molecules that reshape interactions and promote targeted turnover of proteins. Ultimately, the dynamics of substrate receptor interchange in CRLs drive the turnover of selected proteins to influence cellular decision-making.

Geranylgeranylacetone promotes human osteosarcoma cell apoptosis by inducing the degradation of PRMT1 through the E3 ubiquitin ligase CHIP

Lucen Jiang et al.

Summary: The study demonstrates that GGA inhibits proliferation and induces apoptosis in human osteosarcoma cells by upregulating Hsp70 and promoting the degradation of PRMT1. This mechanism leads to FAS-triggered cell apoptosis and suggests that targeting PRMT1 may be a potential therapeutic strategy for osteosarcoma patients.

Summary: Osteosarcoma is the most common malignant bone tumor in children and teenagers, with low survival rates in patients with poor treatment response or metastases. Studies have shown increased expression of four USPs in OS tumor cells, with USP6 and USP41 expression correlated with patient survival. In vivo experiments with a USP inhibitor demonstrated reduced tumor growth and metastases in a preclinical OS model, while in vitro experiments showed decreased cell viability in OS cells through apoptosis induction.
