Variable roles of interleukin-17F in different cancers

Tiina Mikkola et al.

Summary: IL-17F plays a dual role in cancer, with both anti-tumorigenic and pro-tumorigenic effects depending on cancer type and the molecular form and location of IL-17F. Its effects are proposed to be based on multiple mechanisms, such as inhibition of angiogenesis, vasculogenic mimicry, and cancer cell proliferation, migration, and invasion, as well as aggravating the inflammatory process. However, there is no solid evidence for the correlation between IL-17F polymorphisms and cancer incidence or patients' prognosis.

An Immunosenescence-Related Gene Signature to Evaluate the Prognosis, Immunotherapeutic Response, and Cisplatin Sensitivity of Bladder Cancer

Ranran Zhou et al.

Summary: This study analyzed immunosenescence-related genes in bladder cancer and constructed an 8-gene pair model to evaluate prognosis. The model showed predictive value for prognosis, immunotherapeutic response, and cisplatin sensitivity in bladder cancer, which was validated in multiple cohorts and analyses.

A Four-Cell-Senescence-Regulator-Gene Prognostic Index Verified by Genome-Wide CRISPR Can Depict the Tumor Microenvironment and Guide Clinical Treatment of Bladder Cancer

Jian-Xuan Sun et al.

Summary: In this study, senescence genes associated with bladder cancer were identified, and a prognostic index comprising four cell senescence regulator genes was constructed. Significant differences in survival outcomes were observed between high-risk and low-risk groups. Additionally, patients in the high-risk group exhibited higher tumor mutation burden, lower immune scores, higher tumor purity, and better response to immunotherapy and chemotherapy.

Summary: The updated EAU guidelines on UTUC aim to assist clinicians in evidence-based management, providing recommendations on diagnosis, risk stratification, radical and conservative treatment, and prognostic factors. Due to the rarity of UTUC, there is insufficient data to provide strong recommendations. Recommendations are based on the 2017 TNM classification and emphasize individualized treatment based on proposed risk stratification.

Cellular senescence in cancer: from mechanisms to detection

Hui-Ling Ou et al.

Summary: Senescence is a cellular state of stable cell-cycle arrest triggered by stress, involving morphological, intracellular changes and a pro-inflammatory secretory phenotype. It acts as an important tumour-suppressive mechanism, but failure to clear senescent cells may lead to chronic inflammation promoting cancer initiation and progression. Additionally, senescence is a response to post-therapy genotoxic stress, with the emergence of senescent cells being pivotal for detecting potential pro-tumorigenic events.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

MYC as a target for cancer treatment

Michael J. Duffy et al.

Summary: The MYC gene, with its three paralogs, is a common driver gene in cancer, making it an attractive target for cancer treatment. While traditionally considered undruggable, recent advancements have shown potential compounds that can inhibit MYC and exhibit anticancer activity.

Cellular senescence in ageing: from mechanisms to therapeutic opportunities

Raffaella Di Micco et al.

Summary: Cellular senescence is characterized by permanent proliferation arrest and can occur in response to various stresses like telomere dysfunction. Apart from stress-induced senescence, it can also be part of controlled programs in biological processes like embryonic development. Senescent cells contribute to impaired tissue regeneration, chronic diseases, and organismal aging, but also present an opportunity for intervention in aging and associated diseases.

Advances in Immunotherapy and the TGF-β Resistance Pathway in Metastatic Bladder Cancer

David J. Benjamin et al.

Summary: Bladder cancer is a significant burden to global public health, with urothelial carcinoma accounting for nearly 90% of cases. Despite advances in immunotherapy, only a small subset of patients benefit from it. The TGF-beta pathway has been identified as a potential resistance mechanism to immunotherapy, and therapies targeting this pathway are being explored for the treatment of bladder cancer.

Mechanisms of Cellular Senescence: Cell Cycle Arrest and Senescence Associated Secretory Phenotype

Ruchi Kumari et al.

Summary: Cellular senescence is a stable cell cycle arrest triggered in response to various stimuli, characterized by dynamic changes in senescent cells. It has both positive effects on tissue repair and anti-cancer mechanisms, but can also negatively impact organismal health. Multiple pathways, including p53/p21 and p16/pRB, play central roles in regulating this process.

CBX7 suppresses urinary bladder cancer progression via modulating AKR1B10-ERK signaling

Zhengnan Huang et al.

Summary: CBX7 is frequently downregulated in urinary bladder cancer due to promoter hypermethylation, leading to poor prognosis. Its ectopic expression suppresses UBC cell aggressiveness and tumor development by downregulating AKR1B10 and inactivating ERK signaling, providing a potential therapeutic target against UBC.
