Life histories of myeloproliferative neoplasms inferred from phylogenies

Nicholas Williams et al.

Summary: By sequencing the clones of 12 patients with myeloproliferative neoplasms, we identified that early driver mutations are the foundation for tumor development and are linked to lifelong growth and evolution. This discovery provides opportunities for early intervention and a new model for cancer development.

Myeloproliferative Neoplasms: The Long Wait for JAK2-Mutant Clone Expansion

Damien Luque Paz et al.

Summary: Myeloproliferative neoplasms (MPNs) are hematological malignancies caused by somatic mutations from a single HSC. Van Egeren et al. used whole-genome sequencing to reconstruct clonal history and time of acquisition of disease-initiating gene mutation.

Reconstructing the Lineage Histories and Differentiation Trajectories of Individual Cancer Cells in Myeloproliferative Neoplasms

Summary: The JAK2-V617F mutation affects the self-renewal and differentiation dynamics of HSCs in MPNs patients, helping to reconstruct cancer lineages, with the mutation likely occurring several decades before diagnosis and providing a selective advantage for mutant HSCs.
