Empowering Electrochemical Biosensing through Nanostructured or Multifunctional Nucleic Acid or Peptide Biomaterials

Direct electrical contact of NAD+/NADH-dependent dehydrogenase on electrode surface enabled by non-native solid-binding peptide as a molecular binder

Summary: This study introduces a gold-binding peptide to NAD(+)/NADH-dependent formate dehydrogenase (FDH) through genetic modification, enabling direct wiring of the enzyme to the electrode surface. The gold-binding activity of the fused enzymes depends on fusion site and tertiary structure, with the highest activity observed at the C-terminus. The presence of the gold-binding peptide increases the stability of the enzyme electrode and promotes direct electrochemical reactions. This method shows promise for bio-electronic applications involving NAD(+)/NADH-dependent enzymes.

Lithographic Patterning of Nanoscale Arrays of the Oxidase Enzyme CotA: Effects on Activity and Stability

Summary: This study compares the enzymatic activity of nanoscale arrays bearing the oxidase CotA that are produced by two lithographic methods: polymer pen lithography (PPL) and stepper photolithography. The results show that while the photolithography method initially gives higher oxidation activities, the arrays degrade more rapidly compared to those produced by PPL. However, once adjusted for passivation and surface area differences, the immobilized enzymes give essentially the same activity regardless of the lithographic method used.
