Characterization of the mechanism by which a nonsense variant in RYR2 leads to disordered calcium handling

Summary: Heterozygous missense variants of the RYR2 gene can cause catecholaminergic polymorphic ventricular tachycardia. These variants enhance the function of the ryanodine receptors, increasing their sensitivity to calcium activation and leading to an increased propensity for calcium waves and delayed afterdepolarizations. A recent study detected a nonsense variant in RYR2 that resulted in unexplained cardiac arrest. By studying human induced pluripotent stem cells, the researchers found that carvedilol and nebivolol could potentially be effective treatments for the gain of function variants in RYR2.

Whole-exome sequencing reveals a monogenic cause in 56% of individuals with laterality disorders and associated congenital heart defects
