Recent breakthroughs and future directions in drugging aquaporins

Mootaz M. Salman et al.

Summary: Aquaporins play a role in various pathological conditions, and recent research has identified potential routes for inhibiting their function as a new drug development strategy. Developing new drug development frameworks for conditions associated with disrupted water and solute homeostasis will help meet the urgent clinical needs of millions of patients.

Inflammatory Response and Secondary White Matter Damage to the Corpus Callosum after Focal Striatal Stroke in Rats

Rafael Rodrigues Lima et al.

Summary: Stroke, a leading cause of death and disabilities, results in debilitating conditions due to disturbances in cerebral blood vessels. Besides primary metabolic collapse, secondary events like excitotoxicity, inflammatory response, and oxidative stress contribute to additional cell death and functional impairment. This study found that primary ischemic damage in the dorsal striatum can lead to secondary white matter damage in the surrounding corpus callosum in adult rats.

Curcumin mitigates lipopolysaccharide-induced anxiety/depression-like behaviors, blood-brain barrier dysfunction and brain edema by decreasing cerebral oxidative stress in male rats

Sadegh Moradi Vastegani et al.

Summary: This study investigated the anxiolytic and antidepressant-like properties of curcumin and its protective effects on BBB dysfunction and brain edema in LPS-challenged rats. The results showed that curcumin reduced anxiety and depressive-like behaviors, decreased BBB permeability and brain water content, possibly through its antioxidant properties.

Novel Synthetic and Natural Therapies for Traumatic Brain Injury

Denise Battaglini et al.

Summary: Traumatic brain injury (TBI) is a major cause of disability and death worldwide, and the "staircase approach" is considered the only validated treatment method. In addition to conventional therapies, novel neuroprotective therapies have been proposed. Treatment strategies should be based on specific and distinct targets such as brain edema, neuronal activity, anti-inflammatory effects, and more.

Oxidative Stress, Inflammation, and Autophagy: Potential Targets of Mesenchymal Stem Cells-Based Therapies in Ischemic Stroke

Jialin He et al.

Summary: Ischemic stroke is a major cause of death worldwide, with limited treatment options focusing on restoring blood flow; autophagy plays a vital role in brain recovery post-stroke, while dysfunction can worsen cerebral injury; mesenchymal stem cells alleviate oxidative stress and inflammation, regulate autophagy imbalances, and show promise as a treatment for ischemic stroke.

The effects of trifluoperazine on brain edema, aquaporin-4 expression and metabolic markers during the acute phase of stroke using photothrombotic mouse model

Nicole J. Sylvain et al.

Summary: Stroke is the second leading cause of death and the third leading cause of disability worldwide, with edema playing a major role in stroke-associated morbidity and mortality. Regulating water balance in the brain through targeting AQP4 channels may be a promising therapeutic approach for treating brain edema. The FDA-approved drug trifluoperazine (TFP) has shown efficacy in reducing cerebral edema in post-stroke mice by inhibiting AQP4 expression, without inducing deleterious changes in brain electrolytes or metabolic markers.

White Matter Injury After Intracerebral Hemorrhage

Xiongjie Fu et al.

Summary: Spontaneous intracerebral hemorrhage (ICH) is a devastating form of stroke with high morbidity, mortality, and disability rates. Research indicates that the development of white matter injury (WMI) following ICH is complex and determines the prognosis of patients. Comprehensive treatment and understanding the relationship between WMI and other brain cells may reveal therapeutic targets for ICH.
