How cancer cells make and respond to interferon-I

HyeonJoo Cheon et al.

Summary: Acute exposure to high concentrations of IFN-I leads to growth arrest and apoptosis in cancer cells, while chronic exposure to low concentrations provides prosurvival advantages. Phosphorylated ISGF3 drives acute deleterious responses to IFN-I, while unphosphorylated ISGF3 drives essential constitutive prosurvival mechanisms. PD-L1, known for its role in inactivating cytotoxic T cells, also has cell-intrinsic protumor activities, including dampening acute responses to high levels of IFN-I and sustaining expression at low levels that benefit tumors. Understanding the complex roles of IFN-I in cancer may lead to new therapeutic strategies.

Lymphatic Endothelial Markers and Tumor Lymphangiogenesis Assessment in Human Breast Cancer

Jia-Mei Chen et al.

Summary: Metastasis via lymphatic or blood vessels is the leading cause of death for breast cancer. However, the research progress for tumor lymphangiogenesis has lagged behind that for angiogenesis due to the lack of specific markers. With the discovery of lymphatic endothelial cell (LEC) markers, it has been shown that LEC plays an active role in various processes related to lymphatic formation, tumor growth, invasion, and antitumor immunity.

Cancer combination therapies by angiogenesis inhibitors; a comprehensive review

Mohammad Javed Ansari et al.

Summary: Abnormal vasculature is a prominent characteristic of tumor tissue, contributing to tumor immune evasion. Angiogenesis blockade therapy, which aims to disrupt the vascular network and deprive malignant cells of nutrients and oxygen, has been developed. However, cancer cells often develop resistance to anti-angiogenic agents, resulting in enhanced invasiveness and metastasis. This article focuses on the clinical developments of angiogenesis blockade therapy in combination with other conventional treatments, such as immunotherapy, chemoradiotherapy, targeted therapy, and cancer vaccines.

Summary: Angiogenesis plays a vital role in the growth and spread of solid cancers, but breast cancer has shown poor response to anti-angiogenic drugs. Assessing the angiogenic signaling mechanisms in breast cancer, exploring new drug targets, and improving treatment outcomes through vascular normalization and nanotechnology are important areas of research.
