Inclusion of extracellular matrix molecules and necrostatin-1 in the intracapsular environment of alginate-based microcapsules synergistically protects pancreatic β cells against cytokine-induced inflammatory stress

Summary: This study demonstrates that incorporating ECM components and a necroptosis inhibitor into alginate-based microcapsules can reduce the susceptibility of islet cells to inflammatory stress. The combination of collagen type IV/RGD and necrostatin-1 showed higher protective effects on islet cells compared to individual molecules.

Proinflammatory signaling in islet β cells propagates invasion of pathogenic immune cells in autoimmune diabetes

Summary: This study suggests that inflammatory signaling in beta cells promotes autoimmunity during the progression of type 1 diabetes. By deleting a specific proinflammatory gene in mice, the researchers found that the preservation of beta cell mass and reduction in infiltrating T cell populations protected against spontaneous autoimmune diabetes.

Immune cells and their inflammatory mediators modify β cells and cause checkpoint inhibitor-induced diabetes

Summary: Checkpoint inhibitors targeting PD-1/PD-L1 and CTLA-4 have revolutionized cancer treatment but can lead to autoimmune complications. This study found evidence of pancreatic inflammation and shrinkage in patients with CPI-induced diabetes mellitus (CPI-DM). In mouse models, anti-PD-L1 induced diabetes rapidly, while anti-CTLA-4 did not. RNA sequencing revealed infiltrations of cytolytic IFN-gamma(+)CD8(+) T cells in islets with anti-PD-L1. IFN-gamma and TNF-alpha drove changes in beta cells and apoptosis pathways. Inhibition of inflammatory cytokines prevented CPI-DM, providing a new clinical strategy for preventing this complication.

Blocking PD-L1-PD-1 improves senescence surveillance and ageing phenotypes

Summary: The accumulation of senescent cells is a major cause of age-related inflammation and diseases. This study reveals that senescent cells heterogeneously express the immune checkpoint protein PD-L1 and that PD-L1(+) cells accumulate with age. PD-L1(-) cells are sensitive to T-cell surveillance, while PD-L1(+) cells are resistant. The expression of PD-L1 correlates with higher levels of SASP.
