Novel Tools toMeasure SingleMolecules Colocalization in Fluorescence Nanoscopy by Image Cross Correlation Spectroscopy

Simone Pelicci et al.

Summary: Super Resolution Microscopy revolutionized the approach to the study of molecular interactions by providing new quantitative tools to describe the scale below 100 nanometers. We propose new tools based on Image Cross Correlation Spectroscopy (ICCS) to quantify the colocalization of fluorescent signals at single molecule level. The ICCS analysis is an efficient tool to measure relative spatial distribution of different molecular species at the nanoscale.

From Double-Strand Break Recognition to Cell-Cycle Checkpoint Activation: High Content and Resolution Image Cytometry Unmasks 53BP1 Multiple Roles in DNA Damage Response and p53 Action

Laura Furia et al.

Summary: 53BP1 protein plays an important role in sustaining the activity of the p53-regulated transcriptional program. This study uses an automated-microscopy protocol to analyze the evolution of the DNA-Damage Response (DDR) and demonstrates the movement of 53BP1 protein from damaged sites to the nucleoplasm, where it interacts with p53 and enhances the transcriptional regulation of the guardian of the genome protein.

Automated multimodal fluorescence microscopy for hyperplex spatial-proteomics: Coupling microfluidic-based immunofluorescence to high resolution, high sensitivity, three-dimensional analysis of histological slides

Laura Furia et al.

Summary: In situ multiplexing analysis and in situ transcriptomics are revolutionary tools for understanding the molecular basis of cancer and advancing personalized medicine. To achieve the best resolution and sensitivity, continuous interplay between different technologies is required. We propose a new experimental pipeline that alternates between automated and manual procedures to optimize the cost-to-benefit ratio in multiplexing experiments.

Cytoskeleton integrity influences XRCC1 and PCNA dynamics at DNA damage

Verena Hurst et al.

Summary: DNA damage induced by 405-nm laser light recruits proteins involved in base excision repair (BER), but perturbing actin or tubulin polymerization in human cells alters the dynamics of BER factors. Depolymerization of cytoplasmic actin may compromise BER efficiency in mammals due to increased levels of nuclear actin and tubulin, linking cytoskeletal integrity to BER.

TIRR inhibits the 53BP1-p53 complex to alter cell-fate programs

Nishita Parnandi et al.

Summary: TIRR inhibits the formation of the 53BP1-p53 complex by preventing the recruitment of 53BP1 to DSBs, leading to aberrant p53 activity and affecting p53-mediated cell-fate programs.

AHNAK controls 53BP1-mediated p53 response by restraining 53BP1 oligomerization and phase separation

Indrajeet Ghodke et al.

Summary: AHNAK functions as a modulator of 53BP1 by controlling its multimerization potential, thereby affecting the activity of the p53 signaling pathway, resulting in different cell responses in cancer cells and non-transformed cells. Loss of AHNAK sensitizes cells to combinatorial cancer treatments by suppressing p53 target gene networks in tumors.

Single-molecule localization microscopy

Mickael Lelek et al.

Summary: Single-molecule localization microscopy (SMLM) is a powerful imaging technique that allows imaging of biological structures at the molecular scale with improved spatial resolution. By computationally localizing individual fluorescent molecules, super-resolution images can be generated, and experimental considerations and computational processing methods are discussed.
