Summary: Genome mining has revealed the biosynthetic potential of actinomycetes and led to the discovery of novel halogenated bioactive compounds. In this study, a screening for halogenase coding genes in Streptomyces strains isolated from fungus-growing ants identified six genes. Mutants were constructed to study the biosynthetic gene clusters and a novel family of halogenated compounds, named colibrimycins, were identified in Streptomyces sp. strain CS147.

Capturing the microbial volatilome: an oft overlooked 'ome'

Analysis of Streptomyces Volatilomes Using Global Molecular Networking Reveals the Presence of Metabolites with Diverse Biological Activities

Summary: This study is the first to analyze Streptomyces volatilomes using the MSHub/GNPS workflow. By combining the MSHub/GNPS method with conventional approaches, more volatile organic compounds (VOCs) were annotated, many of which have antimicrobial, anticancer, and insect behavior-modulating activities. The discovery of plant-associated VOCs suggests that members of the genus have the potential for large-scale production of these compounds by Streptomyces. Plant-associated VOCs with antimicrobial activities, such as 1-octen-3-ol, octanol, and phenylethyl alcohol, may have applications as fumigants. The annotated VOCs that influence insect behavior provide insights into the functions of Streptomyces VOCs in dispersal and nutrient acquisition.

Natural products in antibiotic development: is the success story over?

Summary: Natural product-based antibiotics have been saving countless lives for more than 80 years. However, there is a lack of research and development in antimicrobial drugs to counteract antimicrobial resistance. Most antibiotics on the market today were inspired by natural product classes discovered over 50 years ago. With advanced genomic approaches and cultivation methods, there is potential for the discovery of even more powerful antibiotics. However, the current antibiotic research and development pipeline is moving away from natural product-based programs, and innovative compounds are rare in early stages.

Ammonia Production by Streptomyces Symbionts of Acromyrmex Leaf-Cutting Ants Strongly Inhibits the Fungal Pathogen Escovopsis

Summary: Leaf-cutting ants have a mutualistic symbiosis with their garden fungus, which can be attacked by a specialized pathogenic fungus. Actinomyces symbionts from the ants contribute to protecting the garden fungus by producing ammonia, which inhibits the growth of the pathogenic fungus Escovopsis. This demonstrates that simple ammonia released by symbionts can play a role in controlling pathogen growth in ant colonies.
