Comparative Analysis of Colon Cancer-Derived Fusobacterium nucleatum Subspecies: Inflammation and Colon Tumorigenesis in Murine Models

They cloned and expressed the riboflavin synthase from a pathogenic bacterium called Leptospira kmetyi, and analyzed its crystal structure. Molecular docking and molecular dynamics simulation were conducted to study the potential inhibitors. The findings shed light on the development of new antibacterial drugs for the treatment of leptospirosis and other related infectious diseases.

Summary: Fusobacterium nucleatum is a dominant bacterial species in colorectal cancer tissue and is associated with cancer progression and poorer patient prognosis. The study reveals that the chemotherapeutic drug 5-fluorouracil can inhibit the growth of Fusobacterium nucleatum, but other bacteria like Escherichia coli in the tumor microbiota have resistance to this drug and can reduce its toxicity on sensitive bacteria and human colorectal cancer cells. Exposing patient CRC tumor microbiota to 5-fluorouracil can disrupt the community and deplete drug levels, reducing local drug efficacy.

The findings revealed that Fusobacterium nucleatum, a Gram-negative obligate anaerobe, is enriched in all cancer tissues and promotes colorectal cancer through the upregulation of the G2M checkpoint pathway. These results suggest that F. nucleatum could be a potential therapeutic target for Korean patients with GI tract cancer.
