Improved Prognostic Precision in Uveal Melanoma through a Combined Score of Clinical Stage and Molecular Prognostication

Bioinformatic Analysis Reveals Central Role for Tumor-Infiltrating Immune Cells in Uveal Melanoma Progression

Summary: This study investigated the role of different tumor-infiltrating cell subsets in uveal melanoma (UM) and proposed a prognostic signature based on the negative prognostic role of CD8+ T cells and M0 macrophages, as well as the positive role of neutrophils. The risk score developed showed superior performance in predicting patient prognosis compared to traditional clinical or pathologic characteristics, with molecular pathways related to cancer immunoevasion and metastasis enriched in the high-risk group. The study suggests further exploration of IDO1 and metalloproteinases as potential novel targets for immunotherapy in lymphocyte-rich metastatic UM patients.

Gene set enrichment analysis (GSEA) indicated activation of immune pathways in low-risk UM phenotype.
