Summary: Pyroptosis and pyroptosis-related genes (PRGs) play crucial roles in the prognosis, progression, and immune microenvironment of pancreatic adenocarcinoma (PAAD). A novel PR risk signature was constructed to assess the prognosis of PAAD, improving traditional prognostic models based on TNM-staging. High PR risk suppressed antitumor immunity by reducing levels of CD8 T and NK cells. Toll like receptor 3 (TLR3) was identified as a potential therapeutic target.

Immunotherapy, a potential treatment method, has shown limited progress in pancreatic adenocarcinoma. This review explores the roles of macrophages and neutrophils as prominent immune cell types in the pancreatic tumor environment and how they impact the harsh tumor environment and standard of care therapies. Targeting macrophages and neutrophils could potentially improve outcomes for patients with pancreatic adenocarcinoma.
