Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Development and characterization of a rat brain metastatic tumor model by multiparametric magnetic resonance imaging and histomorphology

Shuncong Wang et al.

Summary: To facilitate the development of new brain metastasis treatment, a clinically relevant animal model with imaging platform was used to induce rhabdomyosarcoma brain metastasis in rats. Multiparametric MRI was used to systematically analyze the characteristics of the metastasis, which revealed distinct MRI features and demonstrated the effectiveness of a vascular disrupting agent in treating the metastasis.

Heterogeneity of Synchronous Lung Metastasis Calls for Risk Stratification and Prognostic Classification: Evidence from a Population-Based Database

Shuncong Wang et al.

Summary: This study examines the prevalence and prognosis of synchronous lung metastasis (sLM) among cancer patients based on the SEER database. The findings reveal disparities in sLM by primary cancer type and clinicodemographic factors, highlighting the need for risk assessment and prognosis classification tools in clinical management.

The blood-tumour barrier in cancer biology and therapy

Patricia S. Steeg

Summary: The blood-brain barrier plays a crucial role in maintaining normal brain function, but modifications in primary brain tumors and brain metastases result in the blood-tumor barrier, further limiting the penetration of therapeutic agents. This challenge highlights the importance of understanding and overcoming the structural and functional complexities of the blood-tumor barrier for improving cancer therapy and patient outcomes.

ATP-binding cassette transporters restrict drug delivery and efficacy against brain tumors even when blood-brain barrier integrity is lost

Mark C. de Gooijer et al.

Summary: This study investigates the impact of a compromised blood-brain barrier (BBB) on drug treatment of intracranial tumors. It found that regardless of the leakiness of the tumor vasculature, ATP-binding cassette transporters are still present and can reduce drug distribution and efficacy in the tumors. Therefore, BBB leakiness does not guarantee unimpeded access of drugs to the tumor, and BBB-penetrable drugs with little to no affinity for efflux transporters are preferred for intracranial tumor treatment.
