Biomimetic manganese-based theranostic nanoplatform for cancer multimodal imaging and twofold immunotherapy

Yuyue Zhao et al.

Summary: We have developed a nanoplatform capable of improving the immunosuppressive tumor microenvironment (TME) by actively targeting it. By camouflaging the surface of gold nanorods (GNRs) with myeloid-derived suppressor cells (MDSCs) membrane, we have achieved targeted adjustment of the TME. This nanoplatform also enables multimodal cancer imaging and dual immunotherapy.

CD24 blockade promotes anti-tumor immunity in oral squamous cell carcinoma

Ke-Long Zou et al.

Summary: The high expression of CD24 in OSCC is associated with poor prognosis and is significantly correlated with CD68. Inhibiting CD24 can delay tumor growth, reduce TAMs number, increase T cell number, and impair tumor proliferation.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Metabolic adaptation in hypoxia and cancer

Felipe Paredes et al.

Summary: The focus of cells' adaptation to hypoxia is the transcription factor HIF1α, which promotes tumor development by affecting cellular metabolism. In addition to hypoxia, metabolic inhibition also influences HIF1α stability and activity, promoting glycolytic metabolism in cancer cells to support growth and proliferation.

MDSC: Markers, development, states, and unaddressed complexity

Samarth Hegde et al.

Summary: MDSCs are a widely discussed biological entity in immunology, commonly describing cells that arise during chronic inflammation with T cell immunosuppressive functions. Currently lacking clear definitions and a unifying framework across pathologies, we propose a framework based on activation signals to classify MDSCs as discrete cell states. Developing this knowledge of myeloid states across pathological conditions may transform how diseases are grouped and treated.

Cancer Cell Metabolism in Hypoxia: Role of HIF-1 as Key Regulator and Therapeutic Target

Vittoria Infantino et al.

Summary: In cancer cells, metabolic reprogramming under low-oxygen conditions is crucial for tumor survival, with HIF-1 playing a pivotal role. Mitochondria are key players in regulating cellular energy and closely interact with HIF-1 to drive metabolic and functional changes in cancer cells.

Oral Cancer Screening: Past, Present, and Future

S. Warnakulasuriya et al.

Summary: Oral cancer is a significant public health issue, affecting young individuals with poor public awareness. Early detection through visual inspection of premalignant lesions before oral cancer develops can reduce mortality. Screening for high-risk groups is cost-effective, but population-based screening lacks sufficient evidence.

Targeting HIF-1 alpha transcriptional activity drives cytotoxic immune effector cells into melanoma and improves combination immunotherapy

Audrey Lequeux et al.

Summary: Inhibition of HIF-1 alpha transcriptional activity can increase levels of NK and CD8(+) T cells in the tumor microenvironment, enhancing the efficacy of cancer immunotherapy. Combining acriflavine can significantly improve the benefits of cancer immunotherapy based on TRP-2 peptide vaccination and anti-PD-1 blocking antibody, while melanoma patients with high CCL5 levels have a higher survival rate.

Mitochondria-targeted and ultrasound-responsive nanoparticles for oxygen and nitric oxide codelivery to reverse immunosuppression and enhance sonodynamic therapy for immune activation

Changwei Ji et al.

Summary: This study developed mitochondria-targeted nanoparticles to co-deliver oxygen and nitric oxide for enhancing sonodynamic therapy and reversing immunosuppression in the tumor microenvironment. The nanoparticles showed promising results in inhibiting tumor growth, promoting immunogenic cell death, and enhancing immune response both in vitro and in vivo. The findings provide a simple strategy to improve cancer immunotherapy by combining sonodynamic therapy with oxygen and nitric oxide delivery.
