In COVID-19, NLRP3 inflammasome genetic variants are associated with critical disease and these effects are partly mediated by the sickness symptom complex: a nomothetic network approach

Michael Maes et al.

Summary: The NLRP3 inflammasome is activated in response to SARS-CoV-2 infection in COVID-19 patients. Acute infections are accompanied by a sickness symptom complex (SSC) that plays a protective role against COVID-19. Increased SSC can protect against severe COVID-19, while factors such as age, gender, and specific gene variants can attenuate this protection.

Effectiveness of COVID-19 booster vaccines against COVID-19-related symptoms, hospitalization and death in England

Nick Andrews et al.

Summary: This study examined the relative and absolute effectiveness of mRNA booster vaccination against COVID-19. The results showed that the booster dose of BNT162b2 or mRNA-1273 had a relative effectiveness ranging from 85% to 95% against symptomatic disease, and an absolute effectiveness ranging from 94% to 97%. For hospitalization or death, the absolute effectiveness of the BNT162b2 booster ranged from 97% to 99% in all age groups. The study provides real-world evidence of significantly increased protection from the booster vaccine dose against mild and severe disease.

High prevalence of an alpha variant lineage with a premature stop codon in ORF7a in Iraq, winter 2020-2021

Nihad A. M. Al-Rashedi et al.

Summary: Since the first reported case of COVID-19 in China, SARS-CoV-2 has been spreading globally. Genomic surveillance plays a crucial role in tracking the emergence, introduction, and spread of new variants. The rapid increase in COVID-19 infections in Iraq during the second wave is attributed to the introduction of concern variants, likely imported from Europe. Molecular epidemiological analysis revealed the dominance of the B.1.1.7 lineage in Iraq, with multiple strains closely related to those from European countries. A notable frequency of stop codon mutation was observed among the B.1.1.7 lineage sequences, but it didn't significantly affect the function of NS7a protein.

Aberrations in the Cross-Talks Among Redox, Nuclear Factor-κB, and Wnt/β-Catenin Pathway Signaling Underpin Myalgic Encephalomyelitis and Chronic Fatigue Syndrome

Michael Maes et al.

Summary: There is evidence that chronic fatigue spectrum disorders (CFAS-Ds), including myalgic encephalomyelitis (ME), chronic fatigue syndrome (CFS), and chronic fatigue with physiosomatic symptoms, are characterized by neuroimmune and neuro-oxidative biomarkers. This study aimed to analyze the protein-protein interaction (PPI) network of CFAS-D and discover the enriched pathways, molecular patterns, and domains. The results revealed a highly connected network involving immune-oxidative-nitrosative and Wnt/beta-catenin subnetworks. The CFAS-D network was found to be associated with detoxification, inflammatory signaling, and dysfunctions in multicellular organismal homeostatic processes.

Summary: This study aims to explore the physio-affective phenome of long coronavirus disease 2019 (LC) and investigate the effects of oxygen saturation (SpO2) and body temperature during the acute phase on LC. The results indicate that lowered SpO2 and increased body temperature during the acute phase, as well as female sex, can predict the physio-affective phenome of LC. Modulation of the NLRP3 inflammasome by Sars-CoV-2 Envelope protein

Mustafa Yalcinkaya et al.

Summary: Although some drugs and vaccines targeting COVID-19 have shown initial success, understanding the underlying disease pathogenesis of SARS-CoV-2 is crucial for further treatment development. Research indicates that the SARS-CoV-2 envelope protein may initially suppress the host NLRP3 inflammasome response to viral RNA, but potentially increase inflammasome responses in later stages of infection. Targeting the SARS-CoV-2 E protein, especially in early infection stages, could be a novel approach to COVID-19 therapy.
