The ups and downs of caloric restriction and fasting: from molecular effects to clinical application

Sebastian J. Hofer et al.

Summary: Age-associated diseases are on the rise, leading to a need for efficient and low-cost methods. Caloric restriction (CR) is effective but implementation can be challenging, giving rise to CR-mimicking interventions. The advantages and disadvantages of these dietary interventions in humans must be carefully considered.

PXR mediates mifepristone-induced hepatomegaly in mice

Xin-peng Yao et al.

Summary: This study demonstrated that high-dose Mifepristone can induce hepatomegaly in mice through PXR-mediated activation of the YAP pathway. The results showed that Mifepristone can activate human PXR in a concentration-dependent manner and promote the nuclear translocation of PXR and YAP, leading to the expression of their target proteins. High-dose Mifepristone failed to induce hepatomegaly in Pxr-knockout mice, highlighting the crucial role of PXR in Mifepristone-induced hepatomegaly.

Fasting improves therapeutic response in hepatocellular carcinoma through p53-dependent metabolic synergism

Jelena Krstic et al.

Summary: Fasting improves the efficacy of sorafenib and sensitizes sorafenib-resistant HCC by preventing the Warburg shift through reduction in glucose and AKT/mTOR signaling. p53 is necessary for the sorafenib-sensitizing effect of fasting.

YAP-TEAD mediates PPAR α-induced hepatomegaly and liver regeneration in mice

Molecular mechanisms of dietary restriction promoting health and longevity

Cara L. Green et al.

Summary: Dietary restriction with adequate nutrition is considered the gold standard for delaying ageing and extending healthspan and lifespan in various species, as it impacts key nutrient-sensing signalling pathways to promote healthy longevity.

The anti-tumor efficacy of 20(S)-protopanaxadiol, an active metabolite of ginseng, according to fasting on hepatocellular carcinoma

Wenzhen Li et al.

Summary: This study found that fasting can enhance the anti-tumor effects of 20(S)-PPD on hepatocellular carcinoma, including inhibition of cell proliferation, promotion of apoptosis, suppression of cell migration and invasion, and induction of cell cycle arrest. In addition, intermittent fasting significantly enhanced the tumor growth inhibition of 20(S)-PPD without obvious side effects.

Liver regeneration: biological and pathological mechanisms and implications

George K. Michalopoulos et al.

Summary: The liver is a unique solid organ that uses regenerative mechanisms to maintain body homeostasis, unlike other solid organs that adjust to tissue loss but do not return to 100% of normal. This review discusses the regenerative pathways of the liver and the clinical implications of liver regeneration after injury.

Constitutive androstane receptor induced-hepatomegaly and liver regeneration is partially via yes-associated protein activation

Intermittent fasting in the prevention and treatment of cancer

Katherine K. Clifton et al.

Summary: Intermittent fasting (IF) has gained popularity as a potential anticarcinogenic strategy, but its effects on cancer patients remain unclear without high-quality randomized clinical trials. More research is needed to determine the risks and benefits of IF for cancer patients before it can be recommended outside of clinical trials.

Effects of intermittent fasting on liver physiology and metabolism in mice

Jianbo Ma et al.

Summary: Intermittent fasting for 12 hours a day for one month significantly reduced liver weight in mice, with minimal impact on body weight. Fasting led to reduced blood glucose levels and increased alkaline phosphatase levels, as well as global elevation of metabolites in the liver. The metabolic molecules affected by fasting are primarily involved in the citric acid cycle and oxidative phosphorylation.
