The AAA plus superfamily: a review of the structural and mechanistic principles of these molecular machines

Yousuf A. Khan et al.

Summary: ATPases associated with diverse cellular activities (AAA+ proteins) are a superfamily of proteins that are found in all domains of life, transducing chemical energy into mechanical energy through ATP hydrolysis. They perform various cellular functions, including protein disassembly, quality control, DNA replication, and viral replication. Understanding the conserved mechanisms of AAA+ proteins is crucial for health and disease.

Cryo-EM structures reveal high-resolution mechanism of a DNA polymerase sliding clamp loader

Christl Gaubitz et al.

Summary: In this study, the structures of Saccharomyces cerevisiae clamp loader Replication Factor C (RFC) bound to its sliding clamp Proliferating Cell Nuclear Antigen (PCNA) were described, revealing the mechanisms of opening and closing the sliding clamp and the exploration of primer-template junction using a base-flipping mechanism. ATP hydrolysis is triggered by interactions with both PCNA and DNA, explaining the switch-like ATPase activity of RFC. This work provides important insights into the DNA replication mechanism and cellular biology.

Mechanism of replication origin melting nucleated by CMG helicase assembly

Jacob S. Lewis et al.

Summary: The activation of eukaryotic origins of replication involves multiple steps, including the loading of MCM helicase, recruitment of firing factors, and formation of CMG. CMG formation leads to DNA unwinding and melting, which is crucial for origin activation and replication.

Identification of fidelity-governing factors in human recombinases DMC1 and RAD51 from cryo-EM structures

Shih-Chi Luo et al.

Summary: RAD51 and DMC1 recombinases catalyse high-fidelity and mismatch tolerant recombination, processes that are indispensable for the maintenance of genomic integrity. The study elucidates the structural difference between RAD51 and DMC1 with regard to mismatch tolerance, showing that key residues in Loop 1 and Loop 2 play a crucial role in determining the fidelity of DNA recombinases.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.

Mechanistic insight into substrate processing and allosteric inhibition of human p97

Man Pan et al.

Summary: A series of high-resolution cryo-EM structures of substrate-bound human p97 AAA ATPase revealed critical conformational changes and 'power-stroke'-like motions in the ATPase rings, shedding light on the central role of p97 in cellular protein homeostasis. The cryo-EM structure of p97 complexed with the inhibitor NMS-873 at a resolution of 2.4 angstrom demonstrated the mechanism of allosteric inhibition by blocking substrate translocation.
