Associations between ayahuasca consumption in naturalistic settings and current alcohol and drug use: Results of a large international cross-sectional survey

Daniel Perkins et al.

Summary: Emerging evidence suggests that consuming ayahuasca in naturalistic settings is associated with lower self-reported current consumption of alcohol and other drugs, especially for those with prior substance use disorders. This association remains significant even after adjusting for religious or social group effects.

Psychedelic medicines for mood disorders: current evidence and clinical considerations

Jerome Sarris et al.

Summary: Recent studies have found that medicinal psychedelics can modulate brain networks, promote neurogenesis and neuroplasticity. Psilocybin and N,N-dimethyltryptamine/ayahuasca have been found to effectively treat depression, while lysergic acid diethylamide has anxiolytic effects. However, the effects of microdosing psychedelics on mood improvement are still controversial.

Low doses of LSD reduce broadband oscillatory power and modulate event-related potentials in healthy adults

Conor H. Murray et al.

Summary: Classical psychedelics like psilocybin and LSD are being investigated for their potential therapeutic effects in psychiatry. While most studies use high doses, there are reports of beneficial effects of microdosing these drugs. This study found that low doses of LSD resulted in cortical desynchronization and reduced ERP amplitudes, similar to effects seen with higher doses of psychedelics.

Pathophysiology of Major Depression by Clinical Stages

Ana Cecilia de Menezes Galvao et al.

Summary: The study found that patients with first episode MDD had higher levels of CAR and SC compared to controls, while patients with treatment-resistant depression had lower levels of SC and CAR but higher levels of mBDNF and CRP than controls. The severity of depressive symptoms and poor sleep quality were associated with low levels of SC and CAR, and high levels of mBDNF. These findings suggest a strong relationship between clinical stages of MDD and changes in relevant biological markers.

Acute dose-dependent effects of lysergic acid diethylamide in a double-blind placebo-controlled study in healthy subjects

Friederike Holze et al.

Summary: This study evaluated the subjective and autonomic effects of different doses of LSD in healthy subjects. It found that LSD displayed dose-proportional pharmacokinetics and dose-dependently induced subjective responses, with a ceiling effect observed for good drug effects at 100 μg. The results suggest that future LSD research may benefit from dose finding based on these findings, and that the full psychedelic effects of LSD are primarily mediated by serotonin 5-HT2A receptor activation.

Psychedelics in Psychiatry: Neuroplastic, Immunomodulatory, and Neurotransmitter Mechanisms

Antonio Inserra et al.

Summary: Psychedelic compounds are emerging as potential novel therapeutics in psychiatry, with their main effects mediated through the modulation of neurotransmitter systems, immunity, and neuroplasticity. Promising results suggest an opportunity for improvement in the treatment of psychiatric disorders, an area that has seen limited advancement in the past 20 years. Studies are underway to separate the psychedelic effects from the therapeutic effects of these compounds, aiming to provide more effective treatment options.

Potential biomarkers of major depression diagnosis and chronicity

Ana Cecilia de Menezes Galvao et al.

Summary: Specific biomarkers (CAR, SC, mBDNF, and PSQI) show potential in evaluating MDD chronicity, but not for diagnosis. These findings can contribute to further research aiming to develop a stronger model for commercial use in psychiatric clinical practice.

Trial of Psilocybin versus Escitalopram for Depression

Robin Carhart-Harris et al.

Summary: The study compared the efficacy of psilocybin and escitalopram in treating patients with major depressive disorder. While there was no significant difference in antidepressant effects between the two drugs at week 6, secondary outcomes generally favored psilocybin over escitalopram, though these results were not adjusted for multiple comparisons. Larger and longer trials are needed to further compare psilocybin with established antidepressants.

Spontaneous and deliberate creative cognition during and after psilocybin exposure

N. L. Mason et al.

Summary: This study demonstrated that psilocybin can enhance creative thinking acutely while decreasing task-based creativity, and can increase the number of novel ideas seven days later. Brain imaging research indicated that the effects of psilocybin on creativity are linked to connectivity within and between networks in the default mode network.

Transcriptional regulation in the rat prefrontal cortex and hippocampus after a single administration of psilocybin

Summary: The study demonstrates that psilocybin rapidly induces gene expression related to neuroplasticity, biased towards the prefrontal cortex, compared to the hippocampus. This provides further evidence for the rapid plasticity-promoting effects of psilocybin.
