Prospects of telomere-to-telomere assembly in barley: Analysis of sequence gaps in the MorexV3 reference genome

Summary: The study analyzed sequence gaps in the barley genome and found that almost all centromeric sequences and 45S ribosomal DNA repeat arrays were missing in the genome assembly, primarily due to assembly breakdown in long stretches of satellite repeats. However, the missing sequences could not fully explain the difference between assembly size and flow cytometric genome size estimates. The prospects of closing these gaps using ultra-long sequence reads were discussed.
