The potential anti-osteoporotic effect of exercise-induced increased preptin level in ovariectomized rats

Summary: This study investigated the effects of estradiol administration and moderate exercise training on osteoporotic changes in ovariectomized rats. The results showed that both estradiol therapy and moderate exercise training significantly improved bone resorption and formation biomarkers, reduced pro-inflammatory cytokines, and increased bone osteopontin expression. These findings suggest that combined estradiol therapy and moderate exercise training may be a promising therapeutic approach for reducing postmenopausal osteoporosis.

This study found that wheel-running exercise improved bone mineral density and bone microarchitecture in mice. Mechanical Stimulation Protects Against Chondrocyte Pyroptosis Through Irisin-Induced Suppression of PI3K/Akt/NF-κB Signal Pathway in Osteoarthritis

Summary: This study demonstrated that exercise-induced Irisin has anti-inflammatory and antipyroptosis effects on osteoarthritis chondrocytes. Moderate-intensity treadmill exercise increased the levels of Irisin, which recovered the expression of collagen II and attenuated the expression of inflammatory markers. Irisin achieved these effects by inhibiting the PI3K/Akt/NF-kappa B signaling pathway and inhibiting the activity of NLRP3/caspase-1. The findings suggest that Irisin could be a potential therapeutic target for osteoarthritis.

Osteocytes regulate bone anabolic response to mechanical loading in male mice via activation of integrin α5

Summary: This study reveals the significant role of osteocytic integrin alpha 5 in the anabolic response of bone to mechanical loading. Deletion of integrin alpha 5 gene results in increased osteocyte apoptosis and reduced anabolic responses to tibial compression, including decreased osteoblasts and bone formation, and increased osteoclasts and bone resorption. This study suggests that integrin alpha 5 could be a potential therapeutic target for bone loss, especially in the elderly population with decreased mechanical sensitivity.

Irisin Rescues Blood-Brain Barrier Permeability following Traumatic Brain Injury and Contributes to the Neuroprotection of Exercise in Traumatic Brain Injury

Summary: Traumatic brain injury (TBI) is a serious condition with high incidence, mortality, and morbidity globally due to brain edema caused by blood-brain barrier (BBB) dysfunction. The study found that irisin levels in CSF were positively correlated with TBI severity and both endurance exercise and exogenous irisin could reduce BBB damage in a mouse model of TBI. Additionally, irisin was shown to improve BBB dysfunction by promoting UCP2 expression on neuronal mitochondrial membrane, thus alleviating inflammatory response and oxidative stress after TBI.

Summary: Exercise can effectively improve bone formation and metabolism in T2DM mice, with downhill running activating the Wnt3a/beta-catenin pathway to enhance osteogenic capacity and improve bone morphology. Inhibition of the Wnt3a/beta-catenin pathway in the bones of T2DM mice leads to reduced bone formation ability and degradation of bone tissue structure.

Summary: Glucocorticoid-induced osteoporosis (GIOP) is the most common secondary osteoporosis characterized by reduced bone formation. Imbalance between adipogenic and osteogenic differentiation in GIOP BM-MSCs, with a key role played by PPAR gamma 2 and SENP3, contributes to enhanced adipogenesis and weakened osteogenesis in this disorder. Further study revealed that SENP3 de-SUMOylates PPAR gamma 2 on K107 site to promote adipogenesis and inhibit osteogenesis in osteoporotic BM-MSCs, suggesting a potential new strategy and target for clinical diagnosis and treatment of GIOP.
