Default Mode Network quantitative diffusion and resting-state functional magnetic resonance imaging correlates in sporadic Creutzfeldt-Jakob disease

Matteo Paoletti et al.

Summary: This study analyzed the grey matter involvement in sporadic Creutzfeldt-Jakob disease (sCJD) using a multi-parametric MRI approach. The results showed that decreased diffusivity was mainly present in the posterior cortical regions of the Default Mode Network (DMN), as well as in temporal areas and deep nuclei. Interestingly, abnormal increased connectivity was found in both ventral and dorsal components of the DMN in sCJD patients.

Subtype Diagnosis of Sporadic Creutzfeldt-Jakob Disease with Diffusion Magnetic Resonance Imaging

Alberto Bizzi et al.

Summary: The study presents the first practical algorithms for the ante mortem diagnosis of sCJD subtypes. MRI diagnosis of subtype is likely to be achievable in early disease stages, allowing for prognostication of clinical course and design of future therapeutic trials.

Regional Differences in Neuroinflammation-Associated Gene Expression in the Brain of Sporadic Creutzfeldt-Jakob Disease Patients

Ausrine Areskeviciute et al.

Summary: The study found regional and sub-regional differences in the expression patterns of neuroinflammation in the frontal cortex and cerebellum of patients with sporadic Creutzfeldt-Jakob disease, indicating a variable neuroinflammatory response. These differences were not influenced by molecular subtypes of the disease, and there may be overlap in biological processes. Therefore, smaller-scale spatial data are needed to explain the observed differential gene expression.

Multimodal MRI staging for tracking progression and clinical-imaging correlation in sporadic Creutzfeldt-Jakob disease

Simone Sacco et al.

Summary: Diffusion imaging is useful for diagnosing sporadic Creutzfeldt-Jakob disease, but tracking disease progression is limited due to non-linear mean diffusivity changes. A model combining mean diffusivity and volume loss (MRI Disease-Staging) helps understand disease development and potential biomarkers. MRI Disease-Staging correlates with more clinical outcomes than Volume or MD alone, providing insight into sCJD progression.

Biomarkers and diagnostic guidelines for sporadic Creutzfeldt-Jakob disease

Peter Hermann et al.

Summary: Sporadic Creutzfeldt-Jakob disease is a fatal neurodegenerative disease caused by misfolded prion proteins. Its diagnosis relies on a combination of neuropsychiatric symptoms, CSF markers, and imaging. The development of assays like RT-QuIC has improved diagnostic accuracy.

Selective vulnerability to atrophy in sporadic Creutzfeldt-Jakob disease

Kyan Younes et al.

Summary: Brain atrophy in sporadic Creutzfeldt-Jakob disease (sCJD) selectively affects specific cortical and subcortical regions, with involvement of regions overlapping with the default mode network (DMN) and insulae, thalami, and right occipital lobe. Structural equation modeling (SEM) shows an effective connectivity model in sCJD, but not in controls. Presence of visual hallucinations correlates with atrophy in specific brain regions.
