Three-Component Stereoselective Enzymatic Synthesis of Amino-Diols and Amino-Polyols

Grayson J. Ford et al.

Summary: This study presents a three-component strategy for the stereoselective enzymatic synthesis of amino-diols and amino-polyols. Through biocatalytic aldol reactions and reductive aminations, stereoselective formation of amino-polyols was achieved without the need for intermediates isolation.

Cyclisations and Strategies for Stereoselective Synthesis of Piperidine Iminosugars

Debashis Dhara et al.

Summary: This personal account focuses on the synthesis of polyhydroxylated piperidines, a subset of compounds within the iminosugar family, using various cyclisation methods and enantiomerically pure precursors to obtain substituted piperidines. The laboratory has contributed cascades based on reductive amination and Huisgen azide-alkene cycloaddition for the synthesis of substituted piperidines with quaternary centres adjacent to nitrogen.

Screening and characterization of a diverse panel of metagenomic imine reductases for biocatalytic reductive amination

James R. Marshall et al.

Summary: Over 300 new imine reductases have been discovered in this study, and a large and diverse panel of 384 enzymes has been produced for screening. A high-throughput screening method has also been developed. Through these methods, imine reductase biocatalysts capable of accepting structurally demanding ketones and amines for excellent synthesis have been identified.

An in vitro-in vivo sequential cascade for the synthesis of iminosugars from aldoses

Justyna Kuska et al.

Summary: This study introduces a chemoenzymatic approach for the synthesis of biologically important iminosugars from readily available aldoses. By combining transaminase-mediated amination of aldoses with selective oxidation of the transaminase products using Gluconobacter oxydans DSM 2003 whole cells, a panel of valuable iminocyclitols is obtained through chemically catalysed deprotection and reduction steps.
