Sarcopenia - Molecular mechanisms and open questions

Petra Wiedmer et al.

Summary: Sarcopenia is a muscle-wasting syndrome characterized by progressive loss of skeletal muscle mass, quality, and strength during normal aging. Patients with sarcopenia mainly suffer from loss of muscle strength, leading to mobility disorders, increased risk of morbidity, and mortality. Various molecular mechanisms, including hormone function, muscle fiber composition, and inflammatory pathways, have been identified as causes of sarcopenia.

Hydrogen sulfide is neuroprotective in Alzheimer's disease by sulfhydrating GSK3β and inhibiting Tau hyperphosphorylation

Daniel Giovinazzo et al.

Summary: Alzheimer's disease is characterized by memory, executive, and motor function deterioration, with neuropathological hallmarks including neurofibrillary tangles, paired helical filaments, and amyloid plaques. Dysregulation of signaling by hydrogen sulfide has been shown during aging and may play a role in preventing hyperphosphorylation of Tau in Alzheimer's disease. Furthermore, administering the H2S donor sodium GYY4137 to mouse models of Alzheimer's disease has been shown to ameliorate motor and cognitive deficits associated with the disease.

Age-Related Changes in Lipidome of Rat Frontal Cortex and Cerebellum Are Partially Reversed by Methionine Restriction Applied in Old Age

Mariona Jove et al.

Summary: This study examines the impact of aging on the lipidome of rat brain regions through chromatographic techniques and mass spectrometry. It found that aging led to minor changes in lipid profiles, with some reversible effects under a Methionine restriction diet. The frontal cortex was more preserved during aging and maintained under diet intervention, whereas the cerebellum exhibited increased monounsaturated and decreased polyunsaturated contents.

Physiologic Responses to Dietary Sulfur Amino Acid Restriction in Mice Are Influenced by Atf4 Status and Biological Sex

William O. Jonsson et al.

Summary: The study investigated the role of ATF4 as a converging point in the integrated stress response during dietary sulfur amino acid restriction (SAAR). The results showed that ATF4 is essential for the sustained production of endogenous hydrogen sulfide and also plays a role in inducing hepatokine fibroblast growth factor 21. Additionally, biological sex was found to be a determinant of the response to dietary SAAR independent of ATF4 status.
