Thermosensitive and tum or microenvironment activated nanotheranostics for the chemodynamic/photothermal therapy of colorectal tumor

Shige Wang et al.

Summary: This research proposes the one-pot preparation of polydopamine (PDA) decorated mesoporous silica nanoparticle (PMSN) for the thermal and tumor micro-environment (TME) responsive colorectal tumor therapy. The PMSN pores were utilized for loading Fe3+. Lauric acid (LA) was selected as a phase-change ligand to coat the surface of Fe3+-loaded PMSN and prevent the leakage of Fe3+. Bovine serum albumin (BSA) was chosen as a stabilizer to provide colloidal stability for the PMSN-Fe-LA-BSA nanopartilces (PMFLB). Under near infrared laser, the light-sensitive PDA generated heat to kill colorectal cancer cells via hyperthermia. The heat induced the phase-change of LA and triggered the release of Fe3+, which reacted with endogenous H2S in the colorectal TME. The transformed Fe3+ into Fe2+ then initiated the Fenton reaction with H2O2, generating hydroxyl radical (•OH). The Fe2+ was converted back to Fe3+, which further reacted with H2S to enhance the chemodynamic therapy of colorectal tumor. The thermosensitive PMFLB that operates in synergy with the colorectal TME offers a new approach for the rational design of multifunctional nano-therapeutic agents.

Glycyl Radical Enzymes and Sulfonate Metabolism in the Microbiome

Yifeng Wei et al.

Summary: Bacteria play important roles in the biogeochemical sulfur cycle by degrading sulfonates to obtain sulfur, carbon, and energy for growth. Cleavage of the inert sulfonate C-S bond involves various enzymes, cofactors, and both oxygen-dependent and oxygen-independent catalytic mechanisms. Recent discoveries in strictly anaerobic bacteria show that sulfonate degradation occurs through O2-sensitive free radical chemistry, catalyzed by glycyl radical enzymes (GREs), enriching our understanding of sulfonate metabolism in the anaerobic biosphere.

Sulfoquinovose is a select nutrient of prominent bacteria and a source of hydrogen sulfide in the human gut

Buck T. Hanson et al.

Summary: The study reveals that sulfoquinovose (SQ) derived from green vegetables serves as an exclusive microbial nutrient in the human gut, metabolized by specific bacteria such as Eubacterium rectale and Bilophila wadsworthia to produce hydrogen sulfide (H2S), impacting host health. This highlights the importance of individual dietary compounds and organosulfur metabolism on microbial activity and suggests potential for precision editing of gut microbiota through dietary and prebiotic interventions.
