Quiescent human glioblastoma cancer stem cells drive tumor initiation, expansion, and recurrence following chemotherapy

Xuanhua P. Xie et al.

Summary: This study confirms the existence of quiescent cancer stem cells in glioblastoma that can evade anti-proliferative therapies. By analyzing the functional characteristics of glioblastomas from genetically engineered mice, essential quiescent stem-like cells that can be directly isolated from tumors were identified. A specific gene expression signature related to quiescent cancer stem cells was found in both primary and recurrent human glioblastomas. These findings provide insights into the failure of conventional treatment and lay the foundation for alternative therapies.

Thy-1-Integrin Interactions in cis and Trans Mediate Distinctive Signaling

Ping Hu et al.

Summary: Thy-1 is a cell surface glycoprotein that plays diverse biological roles in different cell types. It is involved in various diseases through its direct trans- and cis-interaction with integrins. This review summarizes recent progress in understanding Thy-1-integrin interactions and explores potential binding partners within the integrin regulation/signaling paradigm.

Glioblastomas acquire myeloid-affiliated transcriptional programs via epigenetic immunoediting to elicit immune evasion

Ester Gangoso et al.

Summary: In this study, it was found that GBM stem cells acquire the ability to evade immune clearance through epigenetic immunoediting, leading to the formation of an immunosuppressive tumor microenvironment. This process is not driven by genetic selection but by altering the transcriptional and epigenetic profile of GSCs after immune attack.

Interactions between cancer cells and immune cells drive transitions to mesenchymal-like states in glioblastoma

Toshiro Hara et al.

Summary: The mesenchymal subtype of glioblastoma is believed to be influenced by both cancer cell-intrinsic alterations and external cellular interactions, with macrophages inducing a transition of glioblastoma cells into mesenchymal-like states. This transition is mediated by macrophage-derived oncostatin M (OSM) interacting with its receptors on glioblastoma cells, leading to the activation of STAT3. This study also reveals that mesenchymal-like glioblastoma states are associated with increased expression of a mesenchymal program in macrophages and enhanced cytotoxicity of T cells, suggesting potential therapeutic implications.
