CaMKII Mediates TGFβ1-Induced Fibroblasts Activation and Its Cross Talk with Colon Cancer Cells

Wei Chen et al.

Summary: The study found that CaMKII is overexpressed in stromal CAFs in colon cancer tissue. In vitro experiments showed that TGF beta 1 increased CAF markers expression in human colon fibroblasts, but not in CaMKII depletion fibroblasts. Knockdown of CaMKII significantly inhibited TGF beta 1-induced fibroblasts activation and CAF-like differentiation.

Calmodulin/CaMKII-γ mediates prosurvival capability in apicidin-persistent hepatocellular carcinoma cells via ERK1/2/CREB/c-fos signaling pathway

Wei-Chung Hsu et al.

Summary: This study revealed that CaM/CaMKs enhance the survival capability of liver cancer cells exposed to apicidin by activating signaling cascades, promoting tumor growth and inhibiting apoptosis. Inhibition of CaM effectively attenuated these effects, providing a promising target for cancer treatment.

STIM2: Redox-sensor and effector of the (tumor) microenvironment

Albrecht Schwab et al.

Summary: Ion channels and associated proteins act as a critical interface in cancer cells, sensing and transducing physical-chemical cues from the tumor microenvironment to shape cell behavior. In a recent study, Gibhardt et al. demonstrated how STIM2, a key component of the store-operated Ca2+ entry mechanism in melanoma cells, is regulated by redox signaling.

Neuropilin and tolloid-like 2 regulates the progression of osteosarcoma

Summary: NETO2 is overexpressed in osteosarcoma and acts as an oncogene by activating the PI3K/AKT signaling pathway, promoting cell proliferation, invasion, and epithelial-mesenchymal transition.

Dual NADPH oxidases DUOX1 and DUOX2 synthesize NAADP and are necessary for Ca2+ signaling during T cell activation

Feng Gu et al.

Summary: The formation of Ca2+ microdomains during T cell activation is initiated by the production of nicotinic acid adenine dinucleotide phosphate (NAADP) from its reduced form NAADPH. NADPH oxidases NOX and DUOX play crucial roles in this process, with DUOX2 and G6PD catalyzing a redox cycle that rapidly produces and degrades NAADP through NAADPH.

Aberrant Ca2+ signaling by IP3Rs in adipocytes links inflammation to metabolic dysregulation in obesity

Ekin Guney et al.

Summary: Chronic metabolic inflammation, obesity, and diabetes are closely related to altered regulation of the Ca2+ channel inositol trisphosphate receptor (IP3R) in adipocytes. Studies in mice have shown that adipocyte-specific loss of IP3R1/2 protects against adipose tissue inflammation and insulin resistance, indicating that targeting IP3R-mediated Ca2+ homeostasis in adipocytes could be a potential therapeutic approach for metabolic diseases.

NETO2 promotes esophageal cancer progression by inducing proliferation and metastasis via PI3K/AKT and ERK pathway

Jia-cheng Xu et al.

Summary: Our study revealed that high expression of NETO2 in ESCC patients was associated with tumor staging and lymph node metastasis, promoting ESCC progression through enhancing cell proliferation, suppressing apoptosis, and stimulating tumor growth. Additionally, NETO2 was found to play a crucial role in regulating migration, invasion, and epithelial-mesenchymal transition in ESCC cells. Mechanistically, NETO2 activated ERK, PI3k/AKT, and Nrf2 signaling pathways, with Nrf2 identified as a critical downstream mediator responsible for tumorigenesis. This suggests that NETO2 may serve as a potential therapeutic target or prognostic biomarker for ESCC patients.
