Host Restriction Factors Modulating HIV Latency and Replication in Macrophages

Isabel Pagani et al.

Summary: In addition to CD4(+) T lymphocytes, myeloid cells and differentiated macrophages are targets of HIV-1 infection, and there are significant differences in virus replication between the two cell types. Understanding these differences is crucial for evaluating the role of cell-associated restriction factors in preventing virus replication in macrophages and T cells.

The growth of siRNA-based therapeutics: Updated clinical studies

M. May Zhang et al.

Summary: After more than twenty years since the discovery of the natural gene-silencing mechanism of RNA interference, siRNA-based therapeutics have entered the pharmaceutical market with three approved drugs and many others in advanced stages of development. The majority of late-stage candidates are targeted towards rare diseases or patients who have not responded to current treatments, while some have potential for a wider patient population. This review provides an overview of siRNA mechanisms, barriers to delivery, and profiles of approved drugs and candidates in Phase 3 clinical trials.

MiRNA expression profiling in HIV pathogenesis, disease progression and response to treatment: a systematic review

Carlo Giacomo Leo et al.

Summary: miRNAs play crucial roles in gene expression regulation and dysregulation can lead to human diseases. This study systematically analyzed scientific literature to identify miRNAs associated with HIV, highlighting hsa-miR-29 family, hsa-miR-146b-5p, and hsa-miR-150-5p as frequently reported miRNAs.

Plasma MicroRNA Signature Panel Predicts the Immune Response After Antiretroviral Therapy in HIV-Infected Patients

Jun-Nan Lv et al.

Summary: Approximately 10-40% of HIV-infected individuals fail to achieve immune function improvement after ART, increasing the risk of non-AIDS-related diseases. This study identified five upregulated miRNAs in the plasma of immune non-responders, which were negatively correlated with CD4(+) T cell counts. A predictive model using these miRNAs accurately distinguished between immune non-responders and responders, with further mechanistic insights provided on the role of miR-16-5p in CD4(+) T cell function regulation.

The role of miRNAs in the regulation of autophagy in autoimmune diseases

Sina Khodakarimi et al.

Summary: Autoimmune diseases are chronic injuries caused by specific auto-reactive reactions, with most of their etiology not well understood. Autophagy serves as a protective response to maintain homeostasis by recycling and discrediting cell compartments, playing a crucial role in controlling immune homeostasis.MicroRNAs, on the other hand, are endogenous non-coding small RNAs that play important roles in cell processes and immune system function, with dysregulated miRNAs implicated in the pathogenesis of autoimmune diseases and the regulation of autophagy.
