Glutamine-free mammalian expression of recombinant glycoproteins with uniform isotope labeling: an application for NMR analysis of pharmaceutically relevant Fc glycoforms of human immunoglobulin G1

Saeko Yanaka et al.

Summary: Mammalian cells are commonly used for producing recombinant glycoproteins, but the high production costs are a drawback. We developed a cost-saving method by culturing the cells under glutamine-free conditions for uniform isotope labeling.

HDX-MS and MD Simulations Provide Evidence for Stabilization of the IgG1-FcγRIa (CD64a) Immune Complex Through Intermolecular Glycoprotein Bonds

Kyle W. Anderson et al.

Summary: Previous studies have proposed different models for stabilizing the Fc-Fc gamma RI immune complex. This study used multiple experimental methods to investigate the role of glycoprotein interactions between IgG1 glycans and the FG-loop of Fc gamma RIa in complex stabilization. The results showed that both intermolecular and intramolecular glycoprotein bonds contribute to the stability of the complex.

The Fab portion of immunoglobulin G has sites in the CL domain that interact with Fc gamma receptor IIIa

Yuki Yamaguchi et al.

Summary: This study investigates the interaction between IgG and Fc gamma receptor IIIa (FcγRIIIa). The results show that the Fab of IgG facilitates the interaction by slowing dissociation and increasing enthalpy gain. The CL domain of Fab specifically interacts with the D1 and D2 domains of FcγRIIIa. Additionally, antigen binding plays a role in stabilizing the IgG-FcγRIIIa interactions.
