Antibodies to watch in 2022

Helene Kaplon et al.

Summary: This article provides a summary of key events in the development of commercial antibody therapeutics in 2021 and a glimpse into potential events in 2022. The regulatory review process for antibody therapeutics targeting the SARS-CoV-2 coronavirus has been expedited, leading to emergency use authorizations and full approvals in multiple countries. Additionally, the late-stage commercial clinical pipeline for antibody therapeutics, excluding those for COVID-19, has seen significant growth over the past year.

Tyrosine O-sulfation proteoforms affect HIV-1 monoclonal antibody potency

Cindy X. Cai et al.

Summary: This study explores the potential of CAP256V2LS as a drug for HIV-1 prevention. The researchers developed a method to separate and quantify different sulfation proteoforms and found that the fully sulfated proteoform demonstrated the highest antigen binding and neutralization efficiency. The study also highlighted the variable levels of sulfation produced by different cell lines, which can aid in the development of more potent clinical products.

Sulfotyrosine residues: Interaction specificity determinants for extracellular protein-protein interactions

Valley Stewart et al.

Summary: Tyrosine sulfation is a post-translational modification that enhances protein-protein interaction specificity. Sulfotyrosyl residues (sTyrs) play important roles in various biological processes, including high-affinity interactions, viral entry, thrombin interaction, and collagen binding. The lack of discernible binding site consensus makes sTyr an interesting research topic. Improved methods and advanced techniques will contribute to the discovery and understanding of sTyr occurrence and function.

Evolution of Anti-SARS-CoV-2 Therapeutic Antibodies

Juan C. Almagro et al.

Summary: This review examines the characteristics of nine therapeutic antibodies approved for Emergency Use Authorization in response to the COVID-19 pandemic. It highlights the neutralization profile against variants of concern (VOCs), selection methods, somatic mutations, isotype, Fc modifications, and epitopes recognized on the receptor-binding domain (RBD) of SARS-CoV-2. The review concludes that effective therapeutic antibodies bind to an epitope formed by conserved residues critical for the RBD:hACE-2 interaction. The information presented here can aid in the design of more efficacious antibodies for the prevention and treatment of COVID-19 and other infectious diseases.

Rarely Recognized Antibody Diversification in Covid-19 Evolution to Counteract Advanced SARS-CoV-2 Evasion Strategies, and Implications for Prophylactic Treatment

Siguna Mueller

Summary: The ongoing Covid-19 pandemic has highlighted the importance of finding safe and effective ways to combat the virus and understanding the immune response. Neglected antibody diversification mechanisms, such as post-translational modifications like N or O-linked glycosylation, may play a crucial role in disease evolution, but have not received much attention in the context of SARS-CoV-2. Further research on these mechanisms could provide insights into developing more targeted strategies to combat the virus.
