Harnessing big data to characterize immune-related adverse events

Ying Jing et al.

Summary: Immune-checkpoint inhibitors (ICIs) have been associated with immune-related adverse events (irAEs), a unique spectrum of organ-specific inflammatory toxicities. Recent studies have utilized aggregate clinical data, real-world data, and multi-omics data to investigate the mechanisms and clinical presentations of irAEs. This Perspective provides a summary of the knowledge on irAEs obtained through different sources of "big data".

Knowledge Mapping of Immunotherapy for Hepatocellular Carcinoma: A Bibliometric Study

Jianming Shen et al.

Summary: This study conducted a bibliometric analysis on immunotherapy for hepatocellular carcinoma (HCC) to gain a better understanding of the current situation and potential new research directions. The study found that China is the main research country in this field, with the number of articles increasing every year. The current research hotspots include the immunological landscape of liver cancer, screening of eligible populations for immunotherapy, and the clinical application of immune checkpoint inhibitors.

Research Trends and Most Influential Clinical Studies on Anti-PD1/PDL1 Immunotherapy for Cancers: A Bibliometric Analysis

Yanhao Liu et al.

Summary: This study used bibliometric analysis to identify influential clinical studies and research trends in anti-PD1/PDL1 immunotherapy. The most well-studied cancers were lung cancer and melanoma, and the most influential journal in this area was the New England Journal of Medicine. Further research is needed to identify effective predictive biomarkers, clarify the molecular mechanisms of combined therapy, and establish optimal treatment strategies.

Genome and Transcriptome Biomarkers of Response to Immune Checkpoint Inhibitors in Advanced Solid Tumors

Alexandra Pender et al.

Summary: The study investigated the use of WGTA to analyze fresh tumor biopsies from 98 patients with metastatic tumors, revealing that tumor mutation burden, CD8(+) T-cell, and M1-M2 macrophage ratios were significant predictors of treatment response. Combining multiple biomarkers based on WGTA allows for better identification of responders to immune checkpoint inhibitors in a heterogeneous population of advanced and previously treated cancers.

Predictive Biomarkers for Outcomes of Immune Checkpoint Inhibitors (ICIs) in Melanoma: A Systematic Review

Joosje C. Baltussen et al.

Summary: Immune checkpoint inhibitors have significantly improved the survival of melanoma patients, but durable responses are only seen in a minority, highlighting the need for predictive biomarkers. A systematic review of 177 publications investigating 128 unique biomarkers found T-cell inflamed gene expression profiling as a promising biomarker, while noting high heterogeneity and low sample sizes in current studies. Future research is needed to further explore the utility of candidate biomarkers in melanoma patients receiving ICIs.

Immune-Related Adverse Events in PD-1 Treated Melanoma and Impact Upon Anti-Tumor Efficacy: A Real World Analysis

Melissa L. Bastacky et al.

Summary: This study of 190 metastatic melanoma patients treated with single-agent anti-PD-1 ICI therapy found that 60% experienced at least one immune-related adverse event (irAEs), with irAE occurrence significantly associated with response to anti-PD-1 therapy. Patients with multiple irAEs exhibited distinct patterns, and obesity was distinctly associated with irAE development, while age and gender showed no significant association.
