IL-17 and IL-17-producing cells in protection versus pathology

Kingston H. G. Mills

Summary: IL-17 cytokines have both protective and pathological functions in the context of infection and autoimmunity. They promote protective immunity against pathogens but can also drive inflammatory pathology. Dysregulated IL-17 responses can contribute to immunopathology, and IL-17 is implicated in various inflammatory diseases.

Instructive Cues of Thymic T Cell Selection

Magali Irla

Summary: During thymic development, signals provided by specific cells control the selection of T cells, resulting in the generation of a T cell repertoire capable of responding to foreign antigens while tolerating self-antigens. This review summarizes our current understanding of the signals involved in the selection of regulatory T cells and highlights the functional diversity of thymic antigen-presenting cell subsets implicated in T cell selection.

Mutation of the Polyproline Sequence in CD3ε Evidences TCR Signaling Requirements for Differentiation and Function of Pro-Inflammatory Tγδ17 Cells

Aldo Borroto et al.

Summary: T gamma delta 17 cells play a key role in inflammatory and autoimmune diseases. The role of TCR signaling in the generation and function of T gamma delta 17 cells is not fully understood. The intact CD3 epsilon-PRS sequence is required for optimal differentiation and pathogenic function of T gamma delta 17 cells.

Interleukin-17 governs hypoxic adaptation of injured epithelium

Piotr Konieczny et al.

Summary: Mammalian cells activate HIFs to survive in low-oxygen environments, but injury-induced hypoxia is unable to activate HIF1 alpha. However, IL-17A derived from ROR gamma t(+) gamma delta T cells can activate HIF1 alpha. The IL-17A-HIF1 alpha axis promotes glycolysis for wound healing in epithelial cells, and loss of IL-17RC, HIF1 alpha, or blockade of glycolysis disrupts the repair process. These findings highlight the interconnectedness of inflammation, metabolism, and migration programs, as well as the role of immune cell-derived inputs in cellular adaptation to hypoxic stress during repair.

Constrained TCRγδ-associated Syk activity engages PI3K to facilitate thymic development of IL-17A-secreting γδ T cells

Nital Sumaria et al.

Summary: The study demonstrates that constrained Syk activity optimally engages the PI3K-Akt signaling pathway to promote the development of gamma delta(17) cells and maintain the expression of the IL-17 program. Inhibition of PI3K not only hinders the development of gamma delta(17) cells, but also increases the development of a new subset of gamma delta T cells with a type-I IFN gene expression signature.
