The Role of Tumor Necrosis Factor Alpha (TNF-α) in Autoimmune Disease and Current TNF-α Inhibitors in Therapeutics

Dan-in Jang et al.

Summary: TNF-α was originally identified as a factor causing tumor necrosis but has also been recognized as a pathological component of autoimmune diseases. Its signaling pathways play important roles in cellular responses, and inappropriate activation can lead to chronic inflammation and autoimmune diseases. TNF-α inhibitors have been developed as effective therapeutic tools for various autoimmune diseases.

Summary: The study shows that human Tregs respond positively to proinflammatory cytokines, enhancing proliferation without affecting their lineage identity or function, providing a potential method for therapeutic Treg manufacturing.

Tissue-restricted control of established central nervous system autoimmunity by TNF receptor 2-expressing Treg cells

Summary: The study reveals that TNFR2-expressing Treg cells play a crucial role in suppressing autoimmune diseases in the central nervous system during peak disease, maintaining their functionality and sustained expression of suppressive genes. This late effect also provides insights into the treatment outcomes.

Insights into the biology and therapeutic implications of TNF and regulatory T cells

Benoit L. Salomon

Summary: TNF inhibitors have significant therapeutic effects on autoimmune and rheumatic diseases, but limited efficacy and paradoxical disease exacerbation in some patients may be explained by differences in TNF receptor signaling and the diverse effects of TNF on immune cells, including regulatory T cells. Future TNF-targeting drugs may focus on selectively blocking TNF binding to TNFR1 and on TNFR2 agonists to improve treatment outcomes for rheumatic diseases.

Precision Engineering of an Anti-HLA-A2 Chimeric Antigen Receptor in Regulatory T Cells for Transplant Immune Tolerance

Yannick D. Muller et al.

Summary: The study demonstrates that genome-engineered mono-antigen-specific A2-CAR Tregs can localize to HLA-A2-expressing grafts and exhibit antigen-dependent in vivo suppression. These Tregs do not impair the function of islets and can delay graft-versus-host disease. This approach may be applied towards developing precision Treg cell therapies for transplant tolerance.
