Treatment with interleukin-1 blockade therapy can improve clinical symptoms but does not affect the underlying monoclonal gammopathy. This case highlights the potential therapeutic role of Bruton tyrosine kinase inhibitors in Schnitzler syndrome.

The study identified somatic nonsynonymous single-nucleotide variants, complex structural variants of chromosome 2, and the upregulated expression of macrophage migration inhibitory factor (MIF) in patients with TEMPI syndrome. The level of serum MIF was significantly decreased in one patient after treatment with plasma cell-directed therapy, indicating a potential role of MIF in the pathophysiology of TEMPI syndrome.
