CAR T cells produced in vivo to treat cardiac injury

Joel G. Rurik et al.

Summary: This study developed a therapeutic approach to generate antifibrotic CAR T cells in vivo by delivering modified mRNA in T cell-targeted nanoparticles. The efficacy of these in vivo-reprogrammed CAR T cells was evaluated in a mouse model of heart failure.

Rituximab in patients with acute ST-elevation myocardial infarction: an experimental medicine safety study

Tian X. Zhao et al.

Summary: The use of Rituximab in patients with acute ST-elevation myocardial infarction (STEMI) appears to be safe and has a substantial impact on circulating B-cell subsets. These findings provide important new insights on the feasibility and pharmacodynamics of Rituximab in acute STEMI, which will inform further clinical translation of this potential therapy.

Colchicine prevents disease progression in viral myocarditis via modulating the NLRP3 inflammasome in the cardiosplenic axis

Kathleen Pappritz et al.

Summary: This study evaluated the potential of colchicine to improve CVB3-induced myocarditis. The results showed that colchicine can alleviate cardiac damage, reduce inflammation, and improve cardiac function. In addition, colchicine can inhibit the NLRP3 inflammasome pathway, reducing cardiac fibrosis and cell apoptosis.

Novel strategies for the mitigation of cytokine release syndrome induced by T cell engaging therapies with a focus on the use of kinase inhibitors

Gabrielle Leclercq et al.

Summary: T cell engaging therapies redirect T cells towards tumor cells, leading to cytotoxicity. However, this can trigger a release of pro-inflammatory cytokines causing Cytokine Release Syndrome (CRS), which remains a major safety concern. Mitigation strategies are necessary to reduce cytokine release while maintaining efficacy. Tyrosine kinase inhibitors are emerging as a potential strategy for managing CRS symptoms.

Cytokine release syndrome and associated neurotoxicity in cancer immunotherapy

Emma C. Morris et al.

Summary: CAR T cell therapy has revolutionized the field of cancer treatment, but significant toxicities can occur in up to one-third of patients. The most common toxicities include cytokine release syndrome and immune effector cell-associated neurotoxicity syndrome. Understanding their pathophysiology is crucial for developing novel therapeutics for prevention and management.

Myocarditis and inflammatory cardiomyopathy: current evidence and future directions

Carsten Tschoepe et al.

Summary: Inflammatory cardiomyopathy is a disease characterized by inflammatory cell infiltration into the myocardium and is predominantly mediated by viral infection. Despite extensive research, inflammatory cardiomyopathy complicated by left ventricular dysfunction, heart failure or arrhythmia is associated with a poor prognosis. The relative roles of the pathogen, host genomics and environmental factors in disease progression and healing are still under discussion.

Microenvironmental IL-6 inhibits anti-cancer immune responses generated by cytotoxic chemotherapy

Eric H. Bent et al.

Summary: In a mouse model of BCR-ABL(+) B-cell acute lymphoblastic leukemia, chemotherapy-induced anti-cancer immunity is suppressed by the tumor microenvironment through production of cytokine IL-6. IL-6 inhibits the efficacy of immunotherapy and promotes resistance to doxorubicin, impacting CD8(+) T-cell-mediated anti-cancer responses.

Cancer-A Major Cardiac Comorbidity With Implications on Cardiovascular Metabolism

Daniel Finke et al.

Summary: Cardiovascular diseases are influenced by multiple factors, including traditional risk factors like arterial hypertension, smoking, hyperlipidemia, and diabetes. Recently, there has been a growing focus on comorbidities and their impact on cardiovascular metabolism. Cancer biology and the cardiotoxic effects of chemotherapies may also affect the heart.

Immune Cells and Immunotherapy for Cardiac Injury and Repair

Joel G. Rurik et al.

Summary: Cardiac injury remains a major cause of morbidity and mortality worldwide, with a lack of understanding on why the heart fails to fully recover function after acute injury and why progressive heart failure frequently ensues. Recent studies have brought to light the significant contributions of the immune system to normal cardiac function and response to injury, offering promise for new treatment avenues for cardiac disease through immune modifying therapies.

The Role of NLRP3 Inflammasome in Pericarditis

Adolfo G. Mauro et al.

Summary: In patients with chronic pericarditis and a mouse model of pericarditis, an intensification of inflammasome activation was observed, along with the demonstration of pathological features of pericarditis. NLRP3 inflammasome inhibitor, anakinra, and interleukin-1 trap showed significant improvement in pericardial alterations, while colchicine partially improved pericardial inflammation, highlighting the role of inflammasome in both human and mouse models of pericarditis.
