A method using high-resolution mass spectrometry to identify HIV peptides bound on MHC-1 was utilized, resulting in the discovery of 14 MHC-1 bound peptides from two HIV-1 isolates, with three peptides inducing a CD8 T-cell response. Direct infection of naive CD4 cells by HIV isolates allows for the identification of novel peptides that may be missed by alternative epitope mapping strategies.
