Effect of SARS-CoV-2 seropositivity on antigen - specific cytokine and chemokine responses in latent tuberculosis

Immunopathogenic overlap between COVID-19 and tuberculosis identified from transcriptomic meta-analysis and human macrophage infection

Summary: Current and previous tuberculosis (TB) increase the risk of COVID-19 mortality and severe disease. Mechanisms of immunopathogenic interaction between COVID-19 and TB were explored through transcriptomic analysis. Aberrant expression of specific gene signatures was found in active TB and latent TB progressors, correlating with higher COVID-19 risk. Enrichment of specific macrophages was observed in severe COVID-19 and active TB patients, indicating shared disease exacerbation. In vitro validation demonstrated increased SARS-CoV-2 infection in macrophages cultured in the inflammatory milieu of Mtb-infected macrophages.

The results indicate that a high proportion of individuals who previously had SARS-CoV-2 infection may experience long-term complications, including respiratory, neurological, psychological, and skin-related symptoms. Being female, old age, number of chronic complications, long-term medication, length of hospital stay and ICU stay, and duration of acute symptoms may be significant predictors of post-COVID-19 symptoms.

COVID-19 in patients with HIV-1 infection: a single-centre experience in northern Italy

Summary: The study reported a case series of 26 HIV-infected patients with COVID-19 diagnosed at S.Orsola Hospital in Italy. Among them, 19 were male, with a median age of 54 years, and 73% had one or more comorbidities. Only 5 patients with interstitial pneumonia were hospitalized, and there were no ICU admissions or deaths.

New data on genetics, epidemiology, immune responses, disease severity, and therapy are highlighted, informing our understanding of COVID-19 in this vulnerable population.

HIV infection and COVID-19 death: a population-based cohort analysis of UK primary care data and linked national death registrations within the OpenSAFELY platform

Even after adjusting for age, sex, ethnicity, and a range of comorbidities, the elevated risk remained significant, especially among individuals of Black ethnicity. COVID-19 case fatality rate and tuberculosis in a metropolitan setting

COVID-19 in Patients with Active Tuberculosis

While one patient with advanced TB died despite treatment, the other patient successfully recovered from the coinfection despite also having HIV and renal failure. Relationship of SARS-CoV-2-specific CD4 response to COVID-19 severity and impact of HIV-1 and tuberculosis coinfection

Summary: This study found that the attributes of SARS-CoV-2-specific CD4(+) T cells were associated with the severity of COVID-19, with severe cases showing poor polyfunctional potential, reduced proliferation capacity, and enhanced HLA-DR expression. HIV-1 and TB coinfection can alter the SARS-CoV-2 T cell response, with HIV-1 causing CD4(+) T cell depletion and active TB leading to decreased polyfunctional capacity of SARS-CoV-2-specific CD4(+) T cells in COVID-19 patients. Moreover, COVID-19 patients displayed a reduced frequency of Mycobacterium tuberculosis-specific CD4(+) T cells, which may affect TB disease progression.
