Central Role of Macrophages and Nucleic Acid Release in Myasthenia Gravis Thymus

Summary: This study found that there was no IFN-I signature in the blood of patients with AChR-MG, emphasizing that the IFN-I signature is restricted to the MG thymus. Molecules mimicking endogenous dsDNA signalization increased the expression of IFN-beta and alpha-AChR in thymic epithelial cells and the mouse thymus. A significant decrease in thymic macrophages was demonstrated in AChR-MG, which may lead to the overexpression of IFN-beta and the development of AChR-MG.

Twin study reveals non-heritable immune perturbations in multiple sclerosis

Summary: This study investigated the influence of genetic predisposition and environmental factors on the peripheral immune signatures in monozygotic twins discordant for multiple sclerosis (MS). Through data-driven computational tools and high-throughput single-cell technologies, the researchers identified an inflammatory shift in a specific group of immune cells in twins with MS, as well as the emergence of certain hyper-responsive immune cells. They found that the variance in CD25 expression by certain helper T cells with a naive phenotype was largely driven by genetic and shared early environmental influences. However, the expansion of helper T cells in twins with MS, which were also elevated in non-twin patients with MS, appeared to be independent of individual genetic makeup and correlated with disease severity.

Innate Lymphoid Cells in Autoimmune Diseases

Summary: Innate lymphoid cells (ILC) are a group of immune cells similar to T cells in morphology and cytokine profile, but they lack clonally distributed diverse antigen receptors. ILC, especially ILC1, ILC2, and ILC3, express transcription factors and cytokines similar to T helper cells, and play an important role in innate immunity by responding rapidly to pathogens. Recent studies have shown that alterations in the number and function of ILC are associated with autoimmune diseases such as systemic lupus erythematosus, systemic sclerosis, and rheumatoid arthritis.

Single-cell profiling of myasthenia gravis identifies a pathogenic T cell signature

Summary: This study utilized high-dimensional single-cell mass and spectral cytometry to explore the immune dysregulation in Myasthenia gravis (MG) patients, identifying two dysregulated subsets of inflammatory T helper cells that were reduced in the blood of MG patients and inversely correlated with disease severity. Single-cell RNA-Seq reveals transcriptional heterogeneity and immune subtypes associated with disease activity in human myasthenia gravis

Summary: This study used single-cell sequencing to reveal the transcriptomes of individual cells in patients with myasthenia gravis, identifying 13 major cell clusters and 39 cell subgroups. It was found that B cells, CD4(+) T cells, and monocytes showed more heterogeneity in MG patients, with expanded CD4(+) T cells. Further analyses showed that B cells in MG exhibited higher activity towards plasma cell differentiation, CD4(+) T cell subsets were unbalanced, and monocytes showed highly activated inflammatory pathways. Additionally, a disease-relevant subgroup of CD180(-) B cells was discovered, which was associated with disease activity and anti-AChR antibody levels in MG patients.
