SARS-CoV-2 Variants of Concern Hijack IFITM2 for Efficient Replication in Human Lung Cells

Rayhane Nchioua et al.

Summary: It has been found that the current variants of SARS-CoV-2, including the Omicron variant, depend on IFITM proteins, especially IFITM2, for efficient replication. Depletion of endogenous IFITM2 expression significantly prevents productive infection of these variants in human lung cells. This highlights the crucial role of IFITMs in viral transmission and pathogenicity of SARS-CoV-2.

Acquired chemoresistance can lead to increased resistance of pancreatic cancer cells to oncolytic vesicular stomatitis virus

Dakota W. Goad et al.

Summary: Vesicular stomatitis virus (VSV) is a promising oncolytic virus (OV) for the treatment of pancreatic ductal adenocarcinoma (PDAC). However, some PDAC cell lines are resistant to VSV therapy. This study shows that acquired chemoresistance can affect the efficacy of VSV OV therapy, with gemcitabine-resistant PDAC cells becoming more resistant to VSV. The resistance is correlated with upregulated levels of interferon-stimulated genes associated with chemotherapy resistance.

A novel domain within the CIL regulates egress of IFITM3 from the Golgi and reveals a regulatory role of IFITM3 on the secretory pathway

Li Zhong et al.

Summary: This study uncovers the importance of Golgi egress in the biology of IFITM3 and identifies a domain that regulates this process. The accumulation of IFITM3 in the Golgi and the resulting glycoprotein trafficking defects can be relieved by treatment with Amphotericin B or overexpression of v-SNARE, indicating that IFITM3 interferes with membrane fusion processes important for Golgi functionalities. The comparison of glycoprotein trafficking in WT versus IFITMs-KO cells suggests that modulation of the secretory pathway is a novel feature of IFITM proteins.

S-Palmitoylation and Sterol Interactions Mediate Antiviral Specificity of IFITMs

Tandrila Das et al.

Summary: Interferon-induced transmembrane proteins (IFITMs) are important antiviral proteins that play a crucial role in many viral infections. This study reveals that cholesterol can directly interact with S-palmitoylated IFITMs in cells and alter their conformation in membrane bilayers. Furthermore, modulation of S-palmitoylation levels affects the interaction between IFITMs and cholesterol, thereby influencing the antiviral activity against different viruses.

The Polybasic Cleavage Site in SARS-CoV-2 Spike Modulates Viral Sensitivity to Type I Interferon and IFITM2

Summary: The presence of a polybasic cleavage site in the spike protein of SARS-CoV-2 allows the virus to enter cells in a pH-independent manner, mitigating against IFITM-mediated restriction and promoting replication and transmission. Targeted depletion of IFITM2 expression alleviates potent inhibition of SARS-CoV-2 replication by type I IFNs.
