Orthodontic tooth movement-activated sensory neurons contribute to enhancing osteoclast activity and tooth movement through sympathetic nervous signalling

Hisataka Kondo et al.

Summary: The study showed that sensory nerve injury and sympathectomy both suppressed Orthodontic Tooth Movement (OTM) and reduced osteoclast activity in periodontal tissue. Sensory nerve injury inhibited the increase of sensory and sympathetic nerve markers induced by OTM.Sympathectomy did not reduce sensory nerve markers, but suppressed the increase of sympathetic nerve markers. ISO treatment restored the inhibited tooth movement by sensory nerve injury. Ablation of the VMH also suppressed OTM-induced increases in tooth movement and osteoclast activity.

Sympathetic Neurostress Drives Osteoblastic Exosomal MiR-21 Transfer to Disrupt Bone Homeostasis and Promote Osteopenia

Cheng-Hu Hu et al.

Summary: Sympathetic neurostimulation through beta(1/2)-adrenergic receptor signaling triggers the transcription of miR-21 in osteoblasts, which is then transferred to osteoclast progenitors via exosomes to dictate osteoclastogenesis. Pharmacological inhibition of exosome release can suppress this miR-21 transfer and ameliorate bone loss under stress-induced depression conditions.

Neural control of gut homeostasis

Nasser Abdullah et al.

Summary: This study explores the neural control of gut homeostasis and the interactions between primary afferent nociceptors and various cell types within the gut wall, highlighting their roles in immune regulation, host response to pathogens, and modulation of visceral sensitivity. The gut-brain axis is crucial for maintaining gut homeostasis and influencing higher cognitive functions and emotions, with potential implications for neurological and behavioral disorders.

Crosstalk between skeletal and neural tissues is critical for skeletal health

Nethmi Abeynayake et al.

Summary: Emerging evidence highlights the physical and functional association between neural and skeletal systems in forming a neuro-osteogenic network. This communication is crucial for bone skeletal growth, homeostasis, and repair. Disruptions to sympathetic innervation can accelerate conditions like osteoporosis, while various molecular mechanisms mediate communication between bone and neural populations, emphasizing the significant role of cell surface receptors, cytokines, and ligands in regulating skeletal health.

Interleukin-4 assisted calcium-strontium-zinc-phosphate coating induces controllable macrophage polarization and promotes osseointegration on titanium implant

Da-Wang Zhao et al.

Summary: Titanium and its alloys are promising scaffold materials for dental and orthopedic implants due to their ideal mechanical properties and biocompatibility, but host immune response often leads to implant failures. Surface modification, such as the CSZP coating, is crucial for regulating host immune response and promoting bone regeneration. IL-4 delivery can modulate immune response for bone regeneration on the CSZP coating effectively.

Heterotopic ossification in mice overexpressing Bmp2 in Tie2+lineages

Belen Prados et al.

Summary: Bmp2 signaling is crucial for vascular/hematopoietic lineages, with ectopic expression leading to soft tissue bone formation and HO resembling FOP in mice. Transgenic mice with aberrant Bmp2 activity recapitulate HO pathophysiology, providing a model for investigating potential therapies for disorders related to abnormal bone formation.

Bolei Cai et al.

Summary: This study reveals the critical role of neutrophils in bone regeneration, where levels of IL-8 implanted intramuscularly lead to outcomes resembling fracture hematoma, and certain levels induce ectopic endochondral ossification. Neutrophils are pivotal in recruiting and orchestrating immune cells and BMSCs at the initial stage of bone healing.
