Cell-free DNA profiling informs all major complications of hematopoietic cell transplantation

Summary: Allogeneic hematopoietic cell transplantation (HCT) is an effective treatment for hematologic malignancies and immune disorders. This study shows that cell-free DNA (cfDNA) in blood can be used for monitoring posttransplant complications, including graft-versus-host disease (GVHD), infection, relapse of underlying disease, and graft failure. The study demonstrates the potential of cfDNA to improve the care of HCT recipients by enabling earlier detection and better prediction of these complications.

Limitations and opportunities of technologies for the analysis of cell-free DNA in cancer diagnostics

The Review describes state-of-the-art technologies and highlights the need to address biological and technical challenges to improve cancer diagnostics and patient care.

Cell-Free DNA Promotes Inflammation in Patients With Oral Lichen Planus via the STING Pathway

Summary: This study found that the level of cfDNA is elevated in patients with oral lichen planus (OLP), and the STING pathway activated by cfDNA may play a critical role in the pathogenesis of OLP. Treatment with the cationic polymer PAMAM-G3 can reduce the inflammation induced by cfDNA and may be a potential therapeutic strategy for OLP.

NADPH-oxidase 4 gene over-expression in peripheral blood lymphocytes of the schizophrenia patients

Summary: This study found higher levels of 8-oxodG, NOX4, and BCL2 in the peripheral blood lymphocytes of schizophrenic patients compared to healthy controls. There was a positive correlation between DNA damage markers and apoptotic protein levels in the SZ group. Additionally, a small cellular subfraction with high DNA damage and elevated BAX1 levels was found in the healthy control group.

Summary: Pro-oxidant enzyme Nox4 has been found to play a critical role in TGF beta-induced myofibroblast transformation during fibrosis, and this study aims to explore its involvement in breast cancer. The authors targeted Nox4 in breast patient-derived cancer-associated fibroblasts (CAFs) and found that inhibiting Nox4 significantly suppressed tumor growth and metastasis in mice. They also observed an upregulation of Nox4 expression in CAFs isolated from human breast tumors. Moreover, the study revealed that Nox4 promoted autophagy and survival of CAFs through activation of the Nrf2 pathway. Birc5 was identified as a downstream modulator of the Nrf2-mediated pro-survival phenotype. Overall, these findings highlight the importance of the Nox4-Nrf2 pathway in tumorigenesis and metastasis of breast cancer.
