Transferability of genetic risk scores in African populations

Abram B. Kamiza et al.

Summary: A study has found that polygenic scores for lipid traits derived from African American individuals have high predictive value in a South African Zulu cohort, but not in a cohort from Uganda. This highlights the need to improve polygenic predictions in populations of African ancestries.

Glaucoma Genetic Risk Scores in the Million Veteran Program

Andrea R. Waksmunski et al.

Summary: This study assessed the utility of a genetic risk score (GRS) based on 127 variants for primary open-angle glaucoma (POAG) risk stratification in European and African ancestry veterans, as well as its association with documented invasive glaucoma surgery (IGS).

Evaluation of the Cirrus High-Definition OCT Normative Database Probability Codes in a Black American Population

Victoria Addis et al.

Summary: The aim of this study is to evaluate differences in peripapillary RNFL thickness between healthy Black Americans and the Cirrus HD-OCT normative database. The study found structural differences in the RNFL of Black American patients and suggested reevaluating the Cirrus HDOCT color probability codes to accurately assess glaucomatous nerves in patients of African descent.

Learning from Black/African American Participants: Applying the Integrated Behavioral Model to Assess Recruitment Strategies for a Glaucoma Genetic Study

Ava Kikut et al.

Summary: The underrepresentation of African American participants in medical research perpetuates racial health disparities in the US. A phone interview study was conducted with 50 African American adults in Philadelphia who had previously participated in a genetic study of glaucoma. The study findings reveal that strong instrumental attitudes and normative influences, such as learning more about personal health and contributing to future care options for others, influenced participants' decisions to enroll in this community-promoted research program. Additionally, the experiences of racial discrimination in medical research played a continuing role in African Americans' decisions to participate.

Genetic pre-screening for glaucoma in population-based epidemiology: protocol for a double-blind prospective screening study within Lifelines (EyeLife)

Anna Neustaeter et al.

Summary: Early detection of glaucoma is crucial for maintaining patients' eyesight, as vision loss often starts in the periphery. A genetic pre-screening method called EyeLife aims to increase the efficiency of glaucoma screening programs by focusing on high-risk individuals, potentially improving the visual health of middle-aged and elderly people in the long term.

Characteristics of p.Gln368Ter Myocilin Variant and Influence of Polygenic Risk on Glaucoma Penetrance in the UK Biobank

Nazlee Zebardast et al.

Summary: The study revealed that a significant proportion of individuals with MYOC gene mutations have glaucoma, with many cases remaining undiagnosed. Polygenic risk score contributes to increased disease penetrance and severity.

Genome-wide meta-analysis identifies 127 open-angle glaucoma loci with consistent effect across ancestries

Puya Gharahkhani et al.

Summary: The authors conducted a meta-analysis of genome-wide association studies in 34,179 cases of POAG, identifying 44 previously unreported risk loci and mapping effects across multiple ethnicities. Integration of multiple lines of genetic evidence supports the functional relevance of the identified POAG risk loci.

The Role of Genetic Ancestry as a Risk Factor for Primary Open-angle Glaucoma in African Americans

Brian S. Cole et al.

Summary: This study examines the association of autosomal ancestry with POAG risk in a large cohort of self-identified African Americans. The results show that a higher degree of African ancestry is associated with an increased risk of POAG. Additionally, it is found that both known POAG single nucleotide polymorphisms and an omnigenic ancestry effect influence POAG risk.
