Combinatorial therapies for rescuing myotonic dystrophy type 1 skeletal muscle defects

Aymeric Ravel-Chapuis et al.

Summary: Myotonic dystrophy type 1 (DM1) is a multisystemic disorder with no cure. Recent progress has been made in understanding disease mechanisms and developing new therapies, particularly for skeletal muscle defects. This article highlights the potential of activating AMP-activated protein kinase (AMPK) with different approaches in combination therapies.

Summary: Myotonic dystrophy type 1 (DM1) is a multisystemic disorder caused by CUG repeats in DMPK mRNAs, leading to RNA foci formation and abnormal RNA-binding proteins. AMPK activation and exercise have shown beneficial effects on the DM1 pathophysiology.

Summary: This study investigated the impact of resistance training on FGF-21 and myostatin in elderly men. The results demonstrated that a 12-week resistance training significantly reduced both proteins in elderly men with and without T2D, with a larger reduction of myostatin in elderly men without T2D.

Summary: Disruptions to muscle protein turnover and metabolic regulation contribute to muscle wasting during the progression of cancer cachexia. Initiation of cachexia is also associated with decreased physical activity, and exercise can help improve skeletal muscle AMPK signaling in pre-cachectic mice.
